Expression Of HLA-G At The MRNA And Protein Levels In Pre-eclampsia At Plateau Area Of Qinghai

Objective:To investigate the differentiation expression of HLA-G at the mRNA andthe protein levels in serum and placenta of patients with pre-eclampsia and clarify themolecular immunological mechanism of hypertensive disorder complicatingpregnancy（HDCP） in order to establish the bases for effective clinical treatment andprevention of pre-eclampsia in the environment of chronic hypoxia around highaltitude at plateau area of Qinghai province.Methods:The placental tissue and serum sample of30normal pregnant women,30patients with pre-eclampsias were collected. HLA-G mRNA levels in placental tissuewere investigated by real time fluorescence quantitative polymerase chain reaction.HLA-G mRNA levels was indicated by Ct value.HLA-G protein expression inplacental tissue of normal pregnant women and patients with pre-eclampsias weredetected with Western blotting. Serum sample was tested for solubleHLA-G(sHLA-G)protein by using a validated sandwich ELISA.Results:(1)The level of HLA-G mRNA of placenta in pre-eclampsiagroup(1.42±4.92)was significantly lower than that in controlgroup(9.00±9.94,P＜0.01).(2)The level of HLA-G protein of placenta in pre-eclampsiagroup(0.28±0.04)was significantly lower than that in controlgroup(0.93±0.04,P＜0.01).(3)The maternal serum concentration of sHLA-G in pre-eclampsia group(688.34±181.988)ng/L was significantly decreased as compared with those innormal pregnant women(1030.2±238.568ng/L,P＜0.01). Conclusion:The level of HLA-G mRNA and protein expression in placenta and theconcentration of sHLA-G in serum are all decreased in the pre-eclampticpregnants,which may be related to the development of pre-eclampsia. Thetrophoblasts are able to reduce HLA-G protein expression by down regulation ofHLA-G mRNA transcription.The decreasing of HLA-G expression may play animportant role in the development and occurrence of pre-eclampsia at plateau areaof Qinghai province.
