Objective Through the animal model established, the influence of Zhen Bao Pill on rabbit with atherosclerosis was observaed to clarify the effect of anti-atherosclerosis. Using modern molecular biology methods to explore its mechanism in further research .Found the target of anti-atherosclerosis.Expand its range of indications.To provide experimental basis for clinical application.Methods 1. The rabbits model of aroscleroticthe was established by fedding high fat diet, Observed lipids, blood rheology, morphology of aortic.2.Using UV spectrophotometer to detect rabbit serum nitric oxide (NO), superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), catalase (CAT) activity and malondialdehyde (MDA), 3.By real-time quantitative PCR (Real-time PCR) technique to detect Zhen Bao Pills on rabbit liver low density lipoprotein receptor (LDLR), ribozymes aortic nitric oxide (eNOS), oxidized low density lipoprotein receptor ( LOX-1) gene expression. Results 1.Serum lipids four, found that Zhen Bao Pills can lower cholesterol (TC), triglyceride (TG) and low-density lipoprotein (LDL), Compared with model group, there was significant difference (p>0.05).Zhen Bao Pills high and middle dose group and simvastatin were no significant differences (p<0.05).2Histopathological observation showed that the normal arterial wall structure of the normal rabbits without atherosclerotic lesions, no plaque formation. Model group of rabbit aorta endometriosis was rough; a large number of fat yellow-white stripe pattern and plaque was visible. Compared with model group the plaques of Zhen Bao Pills high and midlle doses group were obviously reduced. It indicated that Zhen Bao Pills could prevent rabbit patch formation.3. Zhen Bao Pills on the impact of hemorheology in rabbits. The results showed that all groups fed high fat diet, whole blood viscosity generally increased,The administration of whole blood viscosity and plasma viscosity was significantly lower than model group (P <0.05).(Except the low dose group).4. Zhen Bao Pills could improve the levels of serum NO in rabbits with atherosclerosis, indicating that the Zhen Bao Pills with the protection of rabbit vascular endothelial function. 5. To detect the serum superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-PX) activity and malondialdehyde (MDA) of rabbit, the results showed that high dose Zhen Bao Pills could improve rabbit serum SOD, CAT, GSH-Px activity and lower MDA content, increased the antioxidant capacity.6. Zhen Bao Pills could enhance liver LDLR and aortic eNOS gene expression, inhibition of aortic LOX-1, VCAM-1 gene expression, this might be the anti-atherosclerosis of a molecular mechanism.Conclusion Zhen Bao Pills had the role of anti-atherosclerosis. The mechanism might be related to regulation of blood lipids; improvement of blood rheology; protection of vascular endothelial; enhancement of the antioxidant capacity by improving the vitality of antioxidant enzymes; Increased liver LDLR and aortic eNOS gene expression, inhibit aortic LOX-1 gene expression and so on.This study more fully reflected the traditional Mongolian way of compound, the role of multi-target advantage, and provide a scientific basis for clinical research applications.
