The Antidepressant Effect Of Kongsheng Zhenzhong Pills In Experimental Study

Objective: Although Kong Sheng Zhen Zhong pill interfere in the mice, analysis the order of precedence of the four drugs: Turtle shell, Keel, Polygalaceae, and Acorus Tatarinowii Schott, and discuss the mechanism in order to provide an theoretical basis for the clinical prevention and control of depression.Methods: The 132 female Kunming mice were randomly divided into 11 groups. Kong Sheng Zhen Zhong pill group also split up into 9 groups (each group of 12),Fluoxetine group (12) and blank group (12). Moreover,Kong Sheng Zhen Zhong pill group devised L₉ (3⁴) scheme, three levels were 5g/Kg, 2g/Kg, no (0). 11 groups were poured the drugs into the stomach continuously for 3 weeks; the fluoxetine group is given about 20mg/Kg, the blank group given saline. Each group all pour the drug into the stomach according to the dose of 0.2mg/10mg.Measure the weight of the mice once a week, then adjust dosage based on the weight. After the last gavage, forced swimming for experiment and the tail suspension test, and separately were recorded for 6 minutes, and the immobility time of the forced swimming and tail suspension after the determination about 4 minutes. The mice were killed immediately after the end of the experiment, took the whole brain, measured in brain tissue SOD, MAO levels.Results: weight change: weight was no significant difference among the groups (P>0.05).Tail suspension test result is: the 2th group, the 5th group, the 6th group, and the 7th group of Kong Sheng Zhen Zhong pill compared with the blank group were markedly different (P<0.05); Other Chinese medicine group and the western medicine group compared with the blank group showed no conspicuous difference (P> 0.05); Traditional Chinese medicine and western medicine, Chinese traditional medicine group is no significant differences between groups.(P>0.05). The results of the forced swimming experiment are: the 6th group, the8th group, and the 9th group of Kong Sheng Zhen Zhong pill compared with the blank group revealed significant differences (P<0.05),the 2th group, the 5th group, and the 7th group of Kong Sheng Zhen Zhong pill, and the fluoxetine group compared with the blank group were prominent difference (P<0.01).The results of the determination of SOD: the 1th group, the 3th group, the 5th group, and the 6th group of Kong Sheng Zhen Zhong pill and the fluoxetine group compared with the blank group were significantly different (P<0.05); there were significant difference between the 1th group of Kong Sheng Zhen Zhong pill group and the fluoxetine (P<0.05). MAO assay results are as follows: The 2th group and the 5th group of Kong Sheng Zhen Zhong pill, compared with the blank group, there were significant difference (P<0.05); there were significantly different between 3 groups, 6 groups, 8 groups and the blank group (P<0.01).Conclusion: Kong Sheng Zhen Zhong pill showed no effect on weight, but obviously shorten the immobility time of the forced swimming experiment and tail suspension test, and increase the content of mouse brain tissue SOD and reduce the content of brain MAO, which proved to have a good antidepressants. The priority of four drugs is: Keel,Polygalaceae, Acorus Tatarinowii Schott, Turtle shell. Optimal compatibility dose is 2g/Kg: 2g/Kg: 2g/Kg: 2g/Kg.
