LPIN1 is a member of LPIN family and a key gene to regulate adipocyte differentiation and lipid metabolism. Whereas there exist opposite conclusions on the effect of LPIN1 gene expression on fatty deposition among mice and natural people, so it’s necessary to take a further research on it with other animal models.Direct sequencing was used in this study to detect the polymorphism of LPIN1 gene among 6 strains including Gushi chicken. We observed thirty-six SNPs and five indel variation sites in 5’end of Exon1 and intron-exon boundary and coding region of Exon6. The sites of -308E1、-16E6、1015E6 and 1128E6 distribute more widely in Gushi chickens. Creative restriction site PCR-RFLP PCR was used to test the distribution of viration of -308E1 and 1015E6 in Gushi chickens and their association with traits. The results showed that chickens of genotype TT had significantly higher abdominal fat and abdominal fat rate than those of genotype CT, and had significantly higher triglyceride content than those of genotype CC; Chickens of genotype CC had significantly higher whole net carcass rate than those of genotype CT and genotype TT; Chickens of GCA deletion homozygotic type had significantly higher heart rate than those of heterozygotic type, and had significantly higher blood triglyceride concentration than those of homozygotic insertion type. The distribution of viration of -16E6 and 1128E6 on exon6-intron boundary in Gushi chicken resource population and it’s association with traits were detected by PAGE, and the results showed that chickens of ATTATT homozygotic deletion in -16E6 had significantly higher leg weight, leg muscle weight and whole net carcass rate than those of heterozygotic type, chickens of ATTATT homozygotic insertion type had significantly higher leg weight rate than those of deletion type; Chickens of TGCTTGCTCAGC homozygotic insertion in 1128E6 had significantly lower spleen weight and blood glucose content than those of heterozygotic type.We obtained four haplotypes in 5’region of exon1 through analysis of variation information of exon1 and exon6 using Phase2.0. Two insertion/deletion E1Hap4 of the four haplotypes mainly distributed in Lushi chicken and Leghorn; In exon6,we also obtained four haplotypes,of which three insertion/deletion E6Hap4 mainly distributed in Gushi chicken,Lushi chicken and Leghorn. Combined analysis of polymorphism information of exon1 and exon6 revealed nineteen haplotypes, 67% of which consisted of five haplotypes(Hap1、Hap8、Hap9、Hap11 and Hap18)with frequency over 5%.In general, there existed abundant polymorphisms in LPIN1 both within strains and between stains. Variation of LPIN1 had significant effect on fat -related characters in Gushi chicken resource population.
