Identification of the genes and polymorphisms underlying quantitative traits, and understanding how these genes and polymorphisms affect economic traits, are important for successful marker-assisted selection and more efficient management strategies in commercial cattle populations. One of the lipin protein family, Lipin1 protein affect the body fat is encoded by LPIN1 gene.LPIN1 gene affect fat development by two ways, one is participate the fat development by the Lipin1 protein which is it encoded; another way is inductive effects, inductive the genes that association with fat development.Genetic polymorphism of LPIN1 gene were detected by PCR-SSCP, PCR-RFLP and DNA sequencing in 740 samples from six cattle populations(Caoyuan red cattle, Qinchuan cattle, Jiaxian red cattle, Luxi cattle, Xianan cattle and Nanyng cattle),and association analysis were carried out by SPSS ware. The purposes of study are to discovery polymorphisms and select molecular markers with significant on growth traits in Chinese cattle, to provide genetic information for molecular breeding in beef database. The research results as follows:The results showed that there were 4SNPs in the LPIN1 gene: g.438C>T, g.31735A>G, g.46137A>C and g.65969G>A, and they located in the UTR, the intron 1, exon 6 and intron 15 respectively, of which 46137A>C was a missense mutation. Association analysis of polymorphisms with economic traits in 260 Qinchuan cattle showed that, the individuals with genotype TT are significantly related to Heart depth, and Heart girth at g.438C>T locus (P<0.05); the individuals with genotype GG are significantly related to Heart depth, Backfat thickness and Heart girth at 31735A>G locus (P<0.05); the individuals with genotype AC and CC are significantly related to Backfat thickness, and the genotype CC is significantly related to the Loin muscle area and Backfat thickness at 46137A>C locus (P<0.05); the individuals with genotype AA are significantly related to the Loin muscle area, Heart depth, Heart girth and Backfat thickness (P<0.05), and the individuals with genotype GG are significantly related to Marbling score (P<0.05) at 465969G>A locus respectively. We conclude that LPIN1 gene may be closely related to the meat quality traits, such as Loin muscle area, Backfat thickness and Marbling score, and it could be as a candidate molecular marker utilized in breeding strategy for Qinchuan cattle.
