Head cabbage (Brassica oleracea L.var. Capitata L.),2n=2x=18, or short cabbage, it is avariation form cruciferae brassica cabbage can form a ball. Head cabbage is native to theMediterranean coast, is not form ball annual wild cabbage through long-term cultivation, its wideadaptability, strong resistance, easy cultivation, stable yield, resistance to transport, so it is themain vegetable crops in many countries in the world. Since the16th century was introduced intoour country, after400years of cultural development, has become the main vegetable crops inChina, and its planting area reached more than ten million acres. Common head cabbage leafsurfaces have different degree of wax, is powder blue or blue-green colour, cuticular wax play animportant role in plant, for example prevent water loss and the ultraviolet radiation damage on thesurface of the plant and resistance to plant diseases and insect pests invasion and otherenvironmental stresses. Glossy wax-less mutant is from “98-1030” when we are doing tissueculture experiment, it is wax-less and glossy, veins are green or yellow green, mutations ofglossiness and green degrees higher than the wild type. Called “glossy98-1030”.We researched four aspects of glossy wax-less mutant: surveyed the field performance;definitized the genetic law; observed leaf cuticular wax ultrastructure through SEM; obtainedclosely chain of molecular markers through AFLP marker. Specific research results are as follows:1. Observation field performance of wax-less mutant.Mutant’s leaf epidermis wax is missing, leaf color is glossy green, veins is green, too. theround leaf number is7, medium maturity, The cabbage ball is round. leaves of wild type cabbagehas wax covering, leaf’s color is blue-green, the round leaf number is8, medium maturity. Exceptfor leaf cuticular wax are different, other aspects also show some differences.2. The genetic law of wax-less glossy character of mutantglossy98-1030and98-1030(wild type) as the parent make hybrid combinations, obtain F1,F2. The separation ratio of with wax plant and wax-less plant was2.86:1, through chi-squareseparation ratio consistent with Mendelian heredity of single gene model, the preliminaryjudgment is that head cabbage wax-less character is controlled by a single recessive gene.3. Observation of ultrastructure of the wax-less mutant epicuticular wax on cabbageDifferent growth periods of development situation of epicuticular wax on cabbage leaveswere observed by scanning electron microscope, and types of wax mutant and wild-type cabbageleaves were selected as materials. Results showed that some waxes crystals were found in the wax mutant, the waxes crystals were immature and granular in whole growth period; different structureof waxes crystals were found in the wild-type, granular and flaky were major crystals structure atseedling stage, cylindrical at head-forming stage, sheet and linear at mature stage.4. Molecular markers of wax-less glossy characterMaterials are two parent and F2segregation population, using256pairs of primers to doAFLP molecular marker, get3probably close chain of molecular markers of wax-less genes.respectively is E40M74-D, E55M33-B, E66M47-F, genetic distance to wax-less gene respectivelyis10.6cM,18.3cM and5.8cM.
