Head cabbage (Brassica oleracea L. var. capitata) is one of the most important vegetablesbelonging to Brassica genus in Cruciferae. It is not only an important vegetable in the supply ofvegetables all year-around, but also one widely cultivated in the world wide. The area of planting hasachieved900000hm2in China.The primary role of cuticular waxes which covering head cabbage more or less is to restrictnon-stomatal water loss and protect the plant from pest and disease damage. Glossy waxless cabbagesare mutants of normal wax head cabbages. Studies have shown that the glossy waxless cabbages areshiny green without the wax bloom. The leaves are shiny, crisp and fresh. The leaf veins are kelly orgreen. They are also rich in nutrient components and have exhibited relatively resistant to pests. So ithas important significance for enriching cabbage germplasm resources. There are extensively studies onthe inheritance of glossy waxless character in Brassica oleracea, it demonstrated diversity amongdifferent materials. Nonetheless, there has been no report about the molecular marker study on theglossy waxless character in cabbage by now. In this study, we clarified the inheritance of glossy waxlesscharacter in cabbage10Q-961, the first time to research molecular marker on the character and obtaineda molecular marker that linked to the glossy waxless gene. We also do the morphological observationabout the glossy waxless character. The main results were as follows:1. The morphological observation about the glossy waxless characterSome botanical characters, including maturity, head shape, plant size and disease resistance, showthat10Q-961exhibits obvious difference with10Q-206. The morphological observation on the glossywaxless cabbage10Q-961and the wax cabbage10Q-206was made. Scanning electron micrographsillustrate that10Q-206has dense vesture of wax crystalline on surface and10Q-961has nothing tocover. The number of stomata in10Q-961was less than in10Q-206.2. The inheritance of the glossy waxless character in cabbage10Q-961F1,F2,BC1P1and BC1P2populations were obtained through crossing,selfing and back-crossing bythe glossy waxless cabbage10Q-961(P1)and the wax cabbage10Q-206(P2).The ratio of wax plantsto glossy waxless plants is3.04:1in F2population, and all the plants are wax in BC1P2population.Genetic analysis preliminarily has shown that the glossy waxless character in cabbage10Q-961wascontrolled by one single recessive gene.3. The molecular marker that linked to the glossy waxless gene in cabbage10Q-961In order to find the DNA marker that linked to the glossy waxless gene in cabbage10Q-961, theDNA of parents and F1were used as templates to screen978pairs of EST-SSR primers,2170pairs ofgSSR primers and255pairs of Indel primers. Combining Bulked Segregant Analysis method, we hadobtained one co-dominant gSSR marker Scaffold2324. Then we make individual test for the F2population with456plants. The marker Scaffold2324was linked to the glossy waxless gene incabbage10Q-961with genetic distance of6.5cM.
