Sorghum is the fifth largest food crops,second only to corn,rice,wheat and barley on theword.Sorghum is drought-tolerant,waterlogging tolerant,poor tolerant,salinity tolerant and multipleresistance.It is widely distributed in arid,semi-arid and low-lying waterlogging areas,with winemaking,broom,weaving and other purposes.Sorghum is also important upland crop.It plays aimportant role to stabilize food production and guarantee food supplies.Using SSR molecular markers,119sorghum materials were used for screening the core SSRprimers for building DNA fingerprinting,and analyzing genetic diversity and population structure.The following results:(1) Based on119sorghum accessions,288SSR primers were used to preliminary screen,second screen, finally screen.41core primers evenly distributed10linkage groups weredetermined for establishing DNA fingerprinting profiles in Sorghum, which could amplify clear,stable and polymorphic bands. Those primers used for building DNA fingerprinting and geneticdiversity evaluation.(2) These primers was suitable for in denaturing polyacrylamide gel electrophoresis. A total of193alleles (na) were obtained with2-9alleles per primer (averaging4.7). The polymorphisminformation content (PIC) ranged from0.1359to0.8171(averaging0.5816);(3) The119sorghum materials were divided into two groups (A: Ⅰ, Ⅱ, Ⅲ; B: Ⅰ, Ⅱ, ⅢandⅣ) by UPGMA cluster;(4) Distance-based cluster and model-based cluster were used to analyze genetic structure andrelationship of78sorghum accessions with41SSR makers. These accessions were divided intofour groups;(5) Comparing with the distance-based cluster,the model-based cluster can distinct the geneticrelationship in the groups,it also analyzes genetic basis among the materials.Some materialscontaining the genetic component of other groups.The results showed that78sorghum accessionswere divided into4groups (Ⅰ, Ⅱ, Ⅲ and Ⅳ). Many accessions in each group were overlappedwith the other groups in genetic component. GroupⅠ had the least genetic distance with groupⅡand had the most genetic distance with group Ⅳ. Based on the genetic relationship among fourgroups, those could be simplified into three groups (A: Ⅰ, Ⅱ; B: Ⅲ; C: Ⅳ) or two groups (A: Ⅰ,Ⅱ, Ⅲ; B: Ⅳ).This methods can improve breeding efficiency.
