The mycelia and artificial fruiting bodies of Cordyceps formosana were investigated systematically by using the anamorphic strain RCEF0868of Hirsutella huangshanensis for the first time, mainly including antimicrobial activities of C. formosana by antimicrobial oxford cup assay, composition analysis of mycelia from shake culture (MSKC), mycelia from submerged culture (MSMC) and artificial culture of fruiting bodies of C. formosana (AFCF), the volatile components extracted by the method of simultaneous distillation and extraction (SDE) in mycelia of C. formosana cultured under different culture conditions and AFCF and analyzed with gas chromatography-mass spectrometry (GC-MS), preliminary research on acute toxicity and subacute toxicity of mycelia of C. formosana.The results of antimicrobial test exhibited that6different kinds of fermentation broth of RCEF0868had different inhibitory effect on Staphylococcus aureus, Bacillus thuringiensis and Bacillus subtilis and the inhibition on S. aureus was the highest and had no inhibitory effect on Candida albicans, Escherichia coli and Pseudomonas aeruginosa. The thermal stability of fermentation broth analysis indicated that the antimicrobial composition may be not sensitive to heat. The antibacterial activity of the myclia of C. formosana was continuously enhanced from the eleventh day to the nineteenth day with S. aureus as indicator. Antibacterial effect of different solvent extracts established that the antibiotic active substance was easily soluble in water, and should belong to the polar substance.Composition analysis displayed that there were plenty and similar quantity of crude protein in all samples. The contents of crude fat and polysaccharide in MSKC were similar to those in MSMC, but higher than that in AFCF. MSKC had the highest content of total amino acids (45.34g/100g) while mannitol and cytosine were the highest in MSMC (27.53,2.44mg/g) and ergosterol in AFCF (6.75mg/g). The content of adenosine in MSKC (1.57mg/g) was significantly higher than that in AFCF (1.32mg/g) and MSMC (0.96mg/g). In addition, there were no statistically significant differences in the content of Mg among all the samples. The content of Ca in AFCF was about2and5times higher than that of MSKC and MSMC. Zn and Cu were the highest in AFCF, while the lowest in MSKC, and Fe, Mn, Ni and Se were significantly higher in MSMC than those of MSKC and AFCF. Besides, the content of As was lower than0.20μg/g in MSKC and MSMC, and the same situation to Cd in MSKC and Hg in all samples tested. Pb was not found in AFCF.GC-MS analysis demonstrated that culture conditions had great influence on the content and number of volatile components, but little impact on compound classes which mainly were alcohols, olefins, esters, phenols and ethers. There were17and12volatile components identified in mycelia of liquid and solid culture with the highest content of1-octene-3-alcohl (51.63%) and2-decene-5-lactone (41.13%), respectively.15volatile compounds were identified from AFCF, with1-octene-3-alcohl contained the highest content of37.70%. The common volatile components of3kinds of artificial culture of C. formosana were1-octene-3-alcohol,2-decene-5-lactone, butylated hydroxytoluene and3-tert-butyl-4-hydroxyanisole.The result of acute toxicity test revealed that mycelia powder of C. formosana had no-toxic side effects on mice and the mice gavage of MTD is greater than7.88g/kg.The result of subacute toxicity test testified that mycelia powder of C. formosana had no-toxic side effects and no obvious toxic reaction on each subject index of rats with the maximum test dose of3.94g/kg, orally for30days.
