In this study,we use Submerged Fermentation Fungal of Fructificatio Corioli, Ganoderma, Ganoderma applanatum, Tremella fuciformis Berk, Cordyceps, Gloeostereum incarnatum, Hericium caput-medusae (Bull:Fr.) Pers as material to estract fungi polysaccharide. The fungi Polysaccharide reacted with tyramine (Tyr) to synthesize fungi Polysaccharide-Tyr, then the production we got reacted with fluorescein isothiocyanate (FITC).The experimental study achievements as follows:(1)The research uses water extracting and alcohol sinking to get fungi polysaccharide.uses sevag method to except protein.(2)The fungi Polysaccharide reacted with tyramine (Tyr) to synthesize fungi Polysaccharide-Tyr.lt is better that GAP and HP reacted with tyramine (Tyr).It is poorer that others fungi Polysaccharide reacted with tyramine (Tyr).(3)According to The result of content、 protein and reducing suger of Fungi polysaccharide and the effecter of the fungi Polysaccharide reacted with tyramine (Tyr), Choose GAP to Fluorescent substance labeling(4)Fluorescent substance labeling of polysaccharide from ganoderma applanatum water-soluble polysaccharide. The Polysaccharide extracting from Ganoderma Applanatum Fermentation reacted with tyramine (Tyr) to synthesize GAP-Tyr, then the production we got reacted with fluorescein isothiocyanate (FITC). We studied the single factor effect of the reaction time,temperature, pH, and the amount of tyramine on GAP-Tyr formation. Base on the single factor experiments,a three-factor-three-level experiment design has been developed by Box-Benhken central composite design method. The result of response surface analysis methodology show that the optimum ultrasonic assisted conditions were reaction time7day, temperature60℃and pH8.(5)In vitro experiment,we use MTT assay to screen the GAP and GAP-Try-FITC.The result show that GAP and GAP-Try-FITC could inhibit the growth of SW1116.
