Taking Silybum marianum Gaertn as an object, make factors influencing Callus induction and growth of research, such as pH of the medium, type of explants, exogenous hormones, nutrients, etc.The results are as follow.To study roots, stems, leaves of Silybum marianum Gaertn as explant, vaccination in plant growth regulator combination ratio for MS+NAA1.0mg/L+ZT1.5mg/L+6-BA1.0mg/L solid culture medium,research on different explant callus induction and the growth of the influence. The results indicate that the different explant of callus induction of different effect. Stem and leaf callus callus induction rate is higher, and the growth of the piece is good, the root of callus callus induction rate low and slow growth. The leaf and the stem callus induction organization are the best explant.To study stems of Silybum marianum Gaertn as explant, vaccination in the experiment of growth hormone screening out the best combination NAA1.0mg/L+ratio ZT1.5mg/L+6-BA1.0mg/L, different basic research on the base of the induced injuries of the organization and the influence of growth. The results indicate that the different basic training of callus induction organization effect are:to strengthen differences. MS induction medium effect is best, callus color is better, induction rate; B5induction medium rate is higher, but the callus color is poorer, N6and White induction rate is low.To study Roots, stems, leaves of Silybum marianum Gaertn as explant, vaccination in NAA,6-BA, ZT and their combination of MS solid culture medium. The results show that:used alone three plants in growth regulating substances certain concentration range are callus induction organizations; Use combination hormone and cytokinins, can obtain the callus induction rate is high, the fast growth, good quality callus; combination of culture medium optimal for:MS+NAA1.0mg/L+ZT1.5mg/L+6-BA1.0mg/Lå'ŒMS+NAA0.75mg/L+ZT0.75mg/L; Low concentration of Silybum marianum Gaertn ZT callus induction to promote the role, but the high concentration of ZT callus have inhibition of growth.Vaccination in additional NAA1.0mg/L+ZT1.5mg/L+6-BA1.0mg/L MS solid culture medium, to stem of Silybum marianum Gaertn as explant, Study of medium pH value of Silybum marianum Gaertn the callus induced and the growth of the influence. The results show that medium pH of5-7when all can induce stem produce Silybum marianum Gaertn callus. The culture medium pH in6.5callus growth best callus induction rate is high, with the pH increases, callus color burn, growth under control.To MS+NAA1.0mg/L+ZT1.5mg/L+6-BA1.0mg/L as medium, stems of Silybum marianum Gaertn as explant, Studies three of nutrient elements in the Silybum marianum Gaertn callus induced and the growth of the influence.
