Silybum marianum (L.) Gaertn is composite Silybum marianum (L.) Gaertn genus.Herbs is that one to two years for health. It is originating in southern Europe and north Africa. In northwest China, north China area extensive introduced variety cultivation, Its fruit have medicinal value. The main effect is that detoxification and clearing heat, protect liver, help protect bravery, protect brain, resistance X rays and so on. The main treatment is that acute or chronic inflammation of the liver, cirrhosis, fatty liver, metabolic toxic liver damage fatty liver, liver disease calculus, cholangitis and around the liver and gallbladder infection and etc. In addition, the content of the seed oil is about28%-36%, and it is higher content nitrogen, phosphorus and potassium in plants. Stem leaves in plant contain that is nitrogen1.54%, phosphorus0.33%and crude protein11%. It has the very high nutritional value. And often eat the oil can prevent arteriosclerosis. Seedling period can be used as a vegetable. In the early spring planting edible on open country field. The present paper reported that obtain a large quantity regeneration plant through tissue culture. Among them, Silybum marianum (L.) Gaertn seeds through eight seconds’alcohol sterilization then twenty five minutes HgCl sterilization, after using sterile water flush6times and then vaccination on B5inducing callus. Foster seven days thus get plants of no bacteria. With plants of no bacteria foster medium. First of all, among the1/2MS+NAA1.0+6-BA0.5+ZT0.5, B5+NAA1.0+6-BA0.5+ZT0.5, N6+NAA1.0+6-BA0.5+ZT0.5, improved white mediums+NAA1.0+6-BA0.5+ZT0.5medium. The experimental results show that it is more suitable for indcing callus base on B5and1/2MS than others. Through subculture, The B5+NAA1.0+6-BAO.5+ZT0.5medium was the most appropriate to induce callus. Using the most appropriate induce callus and medium as the basic materials. For subculture of medium and proliferation. In subculture process, adjusting gradually to use the number of hormone and reduce it. And gradually, make the medium characters stable. According to screening medium of gradual stable character as basic material, through hormone treatment guidance medium differentiation again and form for regeneration plant. Through the experiment, The B5+NAA1.0+6-BAO.5medium was used to induce buds and adventitious roots was induced in the B5+NAA1.0 mediums. As a result of callus, bud and root induction process by the influences of external factors. So we studied in the process of in the induction of bud and root occurs External factors for example, Plant hormones, temperature, light and medium value of pH are effect on callus, bud and root induction rate of callus proliferation and bud and root growth. In order to select the most suitable medium. It is found by experiments: Plant hormone NAA,6-BA and ZT for callus induction has obvious promoting effect. In addition to a certain concentration of NAA,6-BA and ZT’s for B5basic culture medium, induction rate are improved. But when the three kinds of hormone concentration is too high or too low, the induction rate decreases, and even produce inhibition. Plant hormone NAA and6-BA for the induction of adventitious buds have apparent stimulative effect. In addition to a certain concentration of NAA and6-BA’s B5basic culture medium, Induction rate are improved. Plant hormone NAA for the induction of adventitious roots have apparent stimulative effect. In addition to a certain concentration of NAA’s B5basic culture medium.Induction rate are improved. But when the concentration of NAA is greater than or less than1.0mg/L, induction rate decreases, and the inhibition of root induction. To sum up, experimental results show, the appropriate concentration of hormone on callus, bud and root induction has apparent stimulative effect.
