Dangshansu Pear is natived to Dangshansu county of Anhui province, which is the largest pear cultivated variety in China. Because of its big, pulp thick, crispy, Juicy, sweet fruit, it is one of the main export volume of varieties in our county. However, in resent years, as the result of the variety degeneration and cultivation mismanagement and other reasons, the content of sclereid in Dangshansu Pear increases and its pulp becomes thick and palate dreggy, which severely affects the taste and quality of Dangshansu Pear. There are studies that suggest that pollination can improve fruit quality of Dangshansu Pear. Therefore, screening appropriate pollinated varieties is of great importance to improve the Dangshansu Pear’s sclereid content. This experiment used different pollination varieties to pollinate with Dangshansu Pear to study the influence that pollination had affected on the pear fruit quality. Using SRAP molecular markers to analysis the genetic relationship between parents, and to make clear the influence of Dangshansu Pear’s sclereid content and other quality caused by the genetic relationship between pollination varieties. Using cDNA-SRAP technology to make differential Expression of the Genes analysis with the fruits of Dangshansu Pear which have been pollinated with different pollination varieties, and to research whether pollination can change Dangshansu Pear’s Maternal gene expression. It provide theoretical and practical basis for selecting excellent pollination varieties, and improving fruit Dangshansu Pear’s sclereid content and fruit quality.The main research conclusions are as follows:1. During Yali, Zisu, Matihuang, Wonhwang, Huashan, Kosui, Shinseiki, Cuiyu, Cuiguan,and Qiyuesu ten pollinated varieties, we choose Yali, Zisu,Matihuang and Cuiyu as the pollinated varieties with Dangshansu Pear, whose the content of sclereid in Dangshansu Pear fruit are lower than that of natural pollination, and fruit pulp exquisite, taste better. Among these different varieties, the content of fruit saccharic acid ratio which are pollinated by Yali, Matihuang and Cuiyu are higher than that of natural pollination, with its sweet pulp and better flavor. Therefore, we choose Yali, Matihuang and Cuiyu to be pollinated varieties in production.2. It exists correlation in the two of the genetic relationship between pollinated varieties and Dangshansu Pear with the content change of sclereid in Dangshansu Pear.Among them, selecting Zisu, Yali or Matihuang which has the more distant genetic relationship with Dangshansu Pear as the pollinated varieties, the content of sclereid in Dangshansu Pear is lower.On the contrary, selecting Kosui, Huashan or Wonhwang which has the more distant genetic relationship with Dangshansu Pear as the pollinated varieties, the content of sclereid in Dangshansu Pear is lower. Therefore,we can choose those which have the more distant genetic relationship with Dangshansu Pear as the pollinated varieties,to reduce content of sclereid in Dangshansu Pear. and to improve the taste.3. Pollination effects gene expression to influence the fruit quality. CDNA-SRAP technology is suitable for Differential Expression of the Genes analysis with Dangshansu Pear. In this test, we screen18higher repeatability, clear ladder amplification and abundant polymorphism pairs form88pairs of prime to amplificate and analysis data by applying cDNA-SRAP.18pairs of primer combinations produces a total of275clear ladders, and the average primer combinations produces15.3ladders.Among them,there are82polymorphism ladders, which is29.8%of the whole ladders. The average primer produces4.6polymorphism ladders, and it shows good repeatability and polymorphism.
