The experiment used ethanol refluxing extraction to exact crude flavonoids fromChrysanthemum, again by the reduced pressure suction filter, vacuum distillation, removethe alcohol distillation to filtrate is no alcohol taste. For another reduced pressure suctionfilter, and then the chloroform extraction, and finally with polyamide columnchromatography, we can get further purification flavonoids from Chrysanthemum.Vacuum freeze-drying get flavonoids powder, qualitative analysis and quantitative analysis.Including qualitative analysis is divided into color reaction and UV spectra scanning.Compared with standard fluid rutin we can determine the extracted from Chrysanthemumis flavonoids substances. Quantitative analysis through standard rutin curve to calculate theflavonoids of coarse extraction yield, fine extraction yield and flavonoids powder purity.To build the model of carbon tetrachloride acute liver damage mice irrigation to thestomach respectively biphenyl double ester and different doses of flavonoids fromChrysanthemum, using LaiShi method to determine the serum alanine amino shift enzyme,aspartic acid amino transfer the yellow enzyme activity method to determine the superoxide dismutase activity liver tissue, the sulfur and benzodiazepines acid method (TBA)was in the liver tissue mda content, and the comparison of each group, histologic changesobserved the liver.The experimental results show that the material extracted from Chrysanthemum,conforms to the nature of flavonoids substances. The coarse extraction rate of flavonoid is5.87%, fine extraction rate is2.13%and the purity of flavonoids powder is87.95%. In theprotection of acute liver damage of mice, each dose group all can reduce the acutechemical liver damage increased activity in mice, serum liver tissue superoxide dismutase(SOD), reduce the content of mda, different degree can improve liver pathologyorganization damage.
