In order to select anti-cold stress Compound Mongolian Medicine and study its regulation and mechanism on cows in winter, done following experiment:used orthogonal experiment method and low-temperature survival time and the cold water swimming time to screening anti-cold compound Mongolian medicine; at laboratory stage, established acute and chronic cold stress model through continuous6℃cold stress0h2h,24h and1h/d intermittent cold stress10d、20d on mice, the animal of clinical research was Holstein cows in the winter, detected enzyme activity, hormone and cytokine levels of serum, immune function and behavior. The main experimental methods and results are as follows:①the life time of mice fedding No.21group of L25(56) and No.5of L9(34) Compound Mongolian Medicine, called Halun-1soup and Halun-2soup, was significantly longer than mice of fedding water under low temperature of-10℃and cold water of10℃(p<0.01).②the result of immune organ index though weighing immune organs, spleen NK cell activity using MTT method and macrophages phagocytic function though engulfing chicken erythrocytes method:Compound Mongolian Medicine can significantly improve the immune organ index, NK cell activity and phagocytic function of macrophages(p<0.05).③The results of flow cytometry experiment:Compound Mongolian Medicine can improve the percentage of CD3+、 CD4+、 CD19+cell of mice (p>0.05), significantly increase CD4+/CD8+ratio of mice(p<0.05), high dose Compound Mongolian Medicine reduce percentage of CD8+cell(p<0.05); Compound Mongolian Medicine can significantly increase percentage of CD14+cell of cows (p<0.05).④Radioimmunoassay results:Compound Mongolian Medicine can increase Cort of mice (p<0.05),high dose Compound Mongolian Medicine increase IL-2of mice under chronic cold stress(p<0.05); Compound Mongolian Medicine can increase IFN-γ、 IgG、 IL-2of cows (p<0.05).⑤The results of detect With microplate reader:Compound Mongolian Medicine lowered CK、 GOT、 AKP of mice and cows (p<0.05).⑥Compound Mongolian Medicine reduce mice behavior of climbing、 dispersed repose、 walk、 observation (p<0.05); Compound Mongolian Medicine increase ruminant time, frequency of defecation and urination of cows (p<0.05)⑦Compound Mongolian Medicine improve average body temperature, average skin temperature and rectal temperature of cows (p>0.05), lower respiratory frequency (p>0.05); high dose Compound Mongolian Medicine increase lactose, cream,milk protein, non-fat solids, milk production(p>0.05). Conclusion:The Halun-1soup and Halun-2soup can significantly improve non-specificity immune function and cellular immune function of mice and cows under cold stress; Compound Mongolian medicine can significantly decrease the damage of cardiac muscle, liver, skeletal muscle, and even kidney of mice and cows under cold stress; Compound Mongolian medicine can significantly reduce disturbed behavior of mice and affect feed behavior of mice and cows under cold stress; Compound Mongolian medicine can can increase resistance of cows under cold stress in winter to a certain extent, not reduce its production performance.
