Gosling plague (GP) is the causative agent of goose parvovirus (GPV), an acute or subacute and septic infection of gosling and Muscovy ducks. It is one of the most seriously infectious diseases endangering the healthy development of the goose industry, with these symptoms of short course, strong infectivity, quick spread, high mortality rate, and mainly infract against the3-20days of age the gosling. Fang dingyi et al firstly discovered the disease in Yangzhou in1956, and then separated the gosling plague virus strains in1961.Some countries in European reported it in succession in1965.In1987,Li maoxiang and et al determined a part of physical and chemical properties of Yangzhou GPV. So far the prevalence of the disease happens annually and varies different degrees, which has resulted in a severe economic loss and cause the extensive attention from scholars.Gosling plague prevention and treatment rely mainly on attenuated vaccine, inactivated vaccine and hyperimmune serum and egg yolk antibody. Hyerimmune serum contains neutralizing anti-bodies, and they will neutralize the virus specificly and rapidly when the body is invaded, so it can play the role as emergency prevention and treatment. Current serum products, process rough, product purity is not high is one of the main reasons of lack of products, so the use of the protein to distinguish between the various characteristics of the target protein and non-target protein, the use of more advanced technology, make the serumproducts to meet higher standards is the only way for the development of serum products of animal origin. Hyperimmune serum is divided into homologous and heterologous, in2009, Wong Man Keung and others prepared hyperimmune serum antibody titer for the1:8anti-gosling plague Sheep immune,which the protection rate of95%. As an antigen on the goose, heterologous serum is easy to cause the rejection, produce antibodies containing a large number of heterologous antibody.In the preparation process of heterologous antibody, the virus can not proliferate in the host body, resulting in low antibody titers and the effects on the immunity. By homologous hyperimmune serum for emergency prevention and treatment of gosling plague become an inevitable trend.The application of this experimental duck embryo has a large number of amplification of GPV, it collects allantoic fluid by centrifugation to remove impurities, formaldehyde inactivated the preperation of antigen of an immune adult geese. When the agar diffusion titer of1:32, the occipital sinus blood separates and prepares the goose anti-gosling plague plasma. The crude extracts were treated with saturated ammonium sulfate on DEAE Sephadex-A50fine to mention the anti-serum to get anti gosling plague antibodies for the goose.After processing by pepsin F (ab’)2fragment and Fc fragment, using by Sephadex the SP the FF column chromatography to remove the Fc fragment of purified F (ab’)2fragment, serum products to achieve higher standards. The results show that the refined F (ab’)2antigen binding fragments in the and titer of1:1280, for the Laboratory emergency prevention and treatment trials the average success treatment of the goslings at farms in Changchun was88.9%higher than the commercially available hyperimmune serum after the diagnosis, the clinical treatment trials showed a better healing rate.
