In order to investigate the effects of lentinan and astragalus polysaccharide on the immune system function,intestinal inflammatory and anti-oxidative damage of gosling infected with GPV,120 healthy gosling were divided into four groups,including control group,astragalus polysaccharide treatment group,lentinan treatment group with an average of 30 gosling for each group.0.3 mL allantoic fluid contained with GPV was injected into the muscle of group M,group I and group II while group C was infected with the same amount of saline.Group I and group II were fed with 0.5 mL solution of lentinan and astragalus polysaccharide twice a day for 15 days.During the 15-day experiment,these four groups were given the same feed.At the three time points of 5 days,10 days and 15 days after the infection,10 gosling were randomly selected to draw out the heart blood and take out the small intestine after euthanasia.The serum of the heart blood were detected using ELISA kit aimed at the concentration of complement,IFN-?,immunoglobulin.The supernatant was centrifuged after grinding the small intestine tissue to examine the immunoglobulin,IL1?,IL-6 and TNF-?.The amount of m RNA expression of IL1?,IL-6 and TNF-? were detected by q PCR.The superoxide dismutase activity,GSH-Px activity and the concentrate of MDA were also detected.The results showed the structure of small intestine tissue and heart in the astragalus polysaccharide treatment group and lentinan treatment group were more completed than that of GPV infection group with a sharp decrease of infiltration of parenchymal cells,a lighter bleeding symptom and a comparatively integrated intestinal villi.The concentration of s Ig A,Ig G and Ig M in astragalus polysaccharide treatment group and lentinan treatment group was remarkably higher than that of GPV infection group(P<0.05)while the concentration of IL1?,IL-6 and TNF-? previously declared as inflammatory factor in astragalus polysaccharide treatment group and lentinan treatment group was extremely lower than that of the GPV infection group(P<0.05).The amount of m RNA expression of inflammatory factor including IL1?,IL-6 and TNF-? of astragalus polysaccharide treatment group and lentinan treatment group were remarkably lower than that of the GPV infection group(P<0.05)while the superoxide dismutase and GSH-Px activity of astragalus polysaccharide treatment group and lentinan treatment group were extremely higher than that of GPV infection group with an opposite trend of MDA(P<0.05).The results showed the astragalus polysaccharide and lentinan can improve the immune system of gosling through the relation of immunoglobulin s Ig A,Ig G and Ig M in serum and the amount of IFN-?.The two mentioned polysaccharides can also fairly relieve the damage of oxidative and inflammatory to protect the intestinal tissue of gosling infected with GPV and the mechanism of action is closely related to the NF-?B pathway.
