Effects of induction were studied to identify the function of ammonia seawater and5-HT in improving maturation of oocyte stripped from scallop, Argopecten irradians Lamarck. The suitable induction parameters were studied. The egg induced were fertilized and cultivated. The all fertilization process were studied through fluorescence microscopy.Oocytes were stripped and treated by ammonia seawater and5-HT in vitro, and then fertilized artificially. Results showed that ammonia seawater can induce Germinal Vesicle Breakdown (GVBD) of scallop oocytes remarkably, but the fertilizing rate was low.5-HT cannot induce GVBD of oocytes, but can induce the maturation of the oocytes already GVBD. Oocytes first induced by ammonia seawater then induced by5-HT showed the best effects, and the fertilizing rate was (36.47±0.87)%.The research can provide support for the genetic breeding and polyploidy breeding of shellfish.Cytological studies on the induced egg showed that the fertilizing rate of induced egg (37.3±1.46)%was much lower than that of normal egg. The developing speed of fertilized induced egg was also lower than that of normally fertilized egg.The fluorescence microscopy study indicated that the induced egg and the normal egg are in the different stage of the first meiosis. The all fertilizing and developing process of the induced egg was almost as the same as that of the normal egg. This research recorded the all development of the fertilized egg and proved the feasibility of this method.
