In the research of drug of anticancer, we have found that cordycepin have multiple pharmacological effects for cancer patients. And the treatment of leukemia drug made by cordycepin has entered Phase Ⅲ clinical trials. Deamination of cordycepin in vivo, which was catalytic by adenosine deaminase (ADA), make the cordycepin loss its basic biological activity and limits its application in clinical practice.We established the ultra-high performance liquid chromatography (UPLC) method of detecting cordycepin.The standard equation was:y=43872x+8655.6, r=0.9945. The linear correlation coefficient linear range of the precision and stability of the experimental RSD were less than1%.We prepared LDH double hydroxide nanomaterials with ion precipitation method. Then cordycepin was inserted into the bilayer of LDH nanomaterials.We studied the Cordycepin/LDH nanomaterials complex materials inhibition effect on HeLa cells. The results showed that cordycepin/LDH nanomaterials complex have a more pronounced inhibition effect of proliferation of HeLa cells than cordycepin. LDH bilayer has a delayed effect on the procedure of cordycepin release, which provides an experimental basis on clinical application.The cordycepin’s inhibition of proliferation on various tumor cells(cervical cancer cells HeLa, breast cancer cell line MCF-7, resistant Doxorubicin breast cancer cells MCF-7/Dox vascular epithelial cells ECV-304, mice liver cancer cell H22, melanoma cells B16) was detected by MTT assay and WST assay. We investigated the aggregation of autophagosome, which happened during the phenomenon of autophagy, by staining the mitochondria and lysosomes. Detected the LC3-II,which was autophagy instructions protein, conversion and degradation of long-cycle protein P62by western blot method.Establelished the Kunming mouse ascites model and solid tumor model of hepatoma H22.Studied the inhibition of different concentrations of cordycepin on H22in vivo. Observed the modality of tumor cell by HE staining. The results showed that:In vitro,cordycepin has effect of inhibition on different tumor cells proliferation in varying degrees and apoptosis characteristic of the treatment group was obvious.Cordycepin/ LDH complexes was formed after cordycepin inserted LDH bilayer and the inhibition of the proliferation of HeLa cells is enhanced.Cordycepin caused the HeLa cells from the aggregation of macrophage protein LC3-Ⅱ conversion of LC3protein no significant role, but can significantly promote the degradation of p62protein.Cordycepin has a significant inhibitory effect of H22cells in solid tumors, with improving the life of a certain degree of ascites tumor in mice.
