Cordyceps Sinensis is a famous Chinese medicinal fungus that has many active ingredients such as cordycepin, cordyceps acid, and cordyceps polysaccharide, and it also has many pharmacological features. In this thesis, cordycepin was extracted from Cordyceps sinensis and the broad-spectrum antimicrobial of the cordycepin was studied. Methods for industrial production were provided in order to substitute the natural Cordyceps Sinensis.Firstly, Cordyceps sinensis qsun-1strains were activated and cultivated to expand, and the dry powder of Cordyceps sinensis qsun-1strains was prepared for experiments. Secondly, extract cordycepin from Cordyceps sinensis using ultrasonic extraction. The feed to solvent ratio, extract solvent, ultrasonic power and extraction time for cell disruption to the strain qsun-1were determined by single factor experiment and orthogonal experiment. Thirdly, cordycepin was isolated and purified using717strong-based anion exchange resin. Besides, a qualitative and quantitative analysis of cordycepin was conducted by UV spectrophotometry. The broad-spectrum antimicrobial of Cordyceps sinensis qsun-1strains was studied in situation that cordycepin was tested drug, a Gram-positive bacterium Staphylococcus aureus, a Gram-negative bacterium Bacillus subtilis and the fungi Candida albicans were tested bacteria.The study results showed that the optimal values of crucial technological parameters afecting the cordycepin extraction amount were as follows:solvent type methanol, ultrasonic power300w, feed to solvent ratio1:100g/mL and extraction time30min. Under such conditions, the cordycepin extraction amount up to9.275mg/g was obtained. A purity of14.200mg/L (cordycepin/cordyceps) and a more than80%of retention rate could be reached with717strong-based anion exchange resin for cordycepin purification. Besides, UV spectrophotometry was developed for the assay of cordycepin and its figures of merit were evaluated. Broad-spectrum antimicrobial test results showed that:the Cordyceps sinensis qsun-1strains had resistance for the Gram-positive bacterium Staphylococcus aureus, a Gram-negative bacterium Bacillus subtilis and the fungus Candida albicans. In a word, Cordyceps sinensis qsun-1strains had broad-spectrum antimicrobial.
