| The study was mainly to probe an efficient way for the propagation of Sinojackia xylocarpa in term of tissue culture, rapid propagation in virto, physiological and chemical property of callus as well as the interaction between endophyte and plant. The leaves of mature Sinojackia xylocarpa trees were taken as explants. Callus of different texture and morphology was induced from media where different kinds and different concentration of hormones were added. In order to understand the mechanism of physiological and biochemical changes characteristics of callus induced above, we investigated the variation of antioxidant enzymatic activity under the effect of different concentration and combination of hormones. Endophyte of Sinojackia xylocarpa were isolated from leaves taken outside. Interaction between endophyte and plant were studied through coexisting experiment. The results showed as follows:Induction of callus from leaves:Occurring time and forming radio of callus were significantly affected by the type and concentration of plant growth regulators. Compact, mass and fast growth callus were obtained by adding2,4-D and0.25-2mg/L KT. Although occurring time was slower than that induced by2,4-D and0.25-2mg/L KT, callus induced by the combination of NAA and6-BA were smallã€dryã€compactã€nearspherical and white in respect of texture and color. Adventitious Roots were observed on the callus induced by the combination of NAA and6-BA. When another planet were taken as matrial for study, we can found that:inductive formation of roots and callus were obtained respectively when added low and high concentration of auxins.Proliferating and rooting of tissue culture seedlings:The optional proliferating buds culture medium from shoots was WPM supplemented with6-BA lmg/L, NAA1mg/L on which it could grow sturdily. The optional rooting culture medium was WPM supplemented with NAA2.0mg/L,6-BA0.05mg/L and AC0.05%on which rooting percentage was80%.The callus which induced by different concentration combinations of2,4-D and KT were taken as materials for measuring enzymatic activity and MDA contents. With2,4-D concentration increased, SOD activity decreased gradually; POD activity remained near constantly; PPO activity first increased and then decreased. MDA content showed an increasing trend along with2,4-D concentration increased. PPO activity were in inverse correlation to the amount of soluble protein and MDA content were in inverse correlation to the activity of SOD.Endophytes which isolated from leaves of Sinojackia xylocarpa were identified from the aspects of morphology and18S-rDNA gene sequence. In order to study the physiological effects of them, tissue culture seedings were inoculated with them. The result showed that11strains were isolated totally,4of them were bacterials and others were all fungals.6of them were sequenced, intercomparison results showed that5of them were Chaetomium globosum,1strain was ascomycete. Coexisting experimental result showed that most of the bacterial and fungi strain had little effect on the growth of plant, preliminary study showed they were all endophyte. H and F which belong to Chaetomium globosum had growth-promoting effects on the tissue culture seedings... |
