With tissue culture as an assistance measure for the first time,combined with 60Co-γradiation treatment,this research has done the preliminary exploration for tissue culture of salvia miltiorrhiza and combine radiation breeding techniques with salvia miltiorrhiza "99-2,99-3,Bj-shh" as testing materials.Below follows the testing results:First,this research focused on the influence of different breeds,different plants growing adjustment material for callus induction and bud initiation with salvia miltiorrhiza spire in spring as explant,studied periodic changing characteristics of callus.Meanwhile,plant regeneration of breed 99-2 was carried out,and the best method for sound seedling and rootage was studied,so quick, efficient and stable tissue culture and propagation technology was set up.Then,with callus of different breeds of salvia miltiorrhiza lamina induced as material,this research carried out radiation treatment for callus by 10 dosages of 60Co-γradial as 5,10,15,20,25,30,40,60,80,100 Gy,studied the growing and physiological index changes of callus of different breeds of salvia miltiorrhiza and then filter the optimum radiation dosage for different breeds of salvia miltiorrhiza,i.e.median lethal dose for callus;radiation treatment was done to callus induced bud differentiation by 7 different dosages as 5,10,15,20,25,30,40 Gy;radiation research was carried out for median lethal dose of callus induced bud differentiation of different breeds of salvia miltiorrhiza and physiological index changes.Below follows the research results:1.The induction differs for callus and bud differentiation of three breeds of salvia miltiorrhiza by different hormones.Callus induction:99-2 addition 2,4-D 2.0 mg/L,6-BA3.0 mg/Land IBA0.1 mg/L, 99-3 addition 2,4-D1.0 mg/L,6-BA0.5 mg/L and IBA0.1 mg/L,Bj-shh addition 2,4-D2.0 mg/L, 6-BA1.0 mg/L and IBA0.5 mg/L. Bud differentiation:99-2,99-3,Bj-shh all addition 6-BA1.0 mg/L+NAA0.5 mg/L.2.Radiation 60Co-γhas obvious anti-growing effect for three breeds of salvia miltiorrhiza callus. When the radiation dose is 40 Gy,all 99-2 callus browning.Median lethal dose is filtered as 14 Gy for 99-2,31Gy for 99-3 and 33 Gy for Bj-shh.3.Radiation has much influence for physiological index of alllus.When the radiation dose is low, SOD enzymatic activity rises and then falls gradually with the radiation dose increasing.99-3 rises gradually with radiation dose increasing;CAT enzymatic activity of three breeds rises and then falls gradually with radiation dose increasing and then cultivation days extending.4.Radial 60Co-γrestrains callus bud differentiation ability of three breeds.Median lethal dose is filtered as 10 Gy for 99-2,35 Gy for 99-3,13 Gy for Bj-shh. 5.Radiation has great influence for physiological index of callus bud differentiation.SOD enzymatic activity first rises and then falls gradually with radiation dose increasing;99-2 POD enzymatic activity falls gradually with dose increasing;99-3 and Bj-shh POD activity showsthe saddle curve; 99-2,99-3 CAT enzymatic activity rises gradually with radiation dose increasing;while Bj-shh falls and then rises.
