Lilium sargentliae Wilson is a bulb flower bulbs Liliaceae Lilium, also a fresh cut flower material with high ornamental value, extremely highly ornamental value, bulb can be edible and medicinal value. With ornamental value、medicinal value, Lilium sargentliae Wilson has the extremely high commercial potential and market prospects, but it is subjected to its lowly fecundity、excessivly collection、the continuous deterioration of the natural environment, it is very difficult to see the wild Lilium sargentliae Wilson in its origin. At present, the researches on the tissue culture of Lilium sargentliae Wilson is single, and has not formed a systematic study and analysis.This paper takes Lilium sargentliae Wilson’s scales, filaments, Bulbilss and the induced aseptic seedling leaves as four kinds of explants, to investigate the propagation of proliferation, differentiation and rooting of plantlets, the key technology and formula of smelting plants respectively, and compared to four kinds of explants results, in order to find the most suitable medium, culture conditions and the most suitable explants for tissue culture of Lilium sargentliae Wilson. The results of the study not only can effectively protect the endangered state of Lilium sargentliae Wilson germplasm resources, but also provide technical reference and scientific basis to its industrialization and commercialization. The results of the study are as follows:1、The Bulbils of Lilium sargentliae Wilson as explant in vitro culture, better Bulbils for callus induction medium is MS+6-BA 1.5 mg/L+NAA 0.15mg/L,the callus induction rate can reach 76.7%; The best bulbil and adventitious Bulbils medium for induction and proliferation is MS+6-BA 1.5mg/L+NAA 0.1 mg/L,Value-added coefficient reach 7.28; Bulbils increment coefficient suitable for adventitious Bulbils rooting medium is 1/2MS+IBA0.5mg/L, the rooting rate up to 88.9%, the average root length 2.4 cm.2、Scales with Lilium sargentliae Wilson as explant in tissue culture, MS+6-BA1.0 mg /L+NAA0.3mg/L is the best callus induction medium; The induced rate reached 68.9%; MS+6-BA 2.0mg/L+NAA 0.1 mg/L is the best of adventitious Bulbils induction and proliferation medium, increment coefficient up to 7.68, and adventitious Bulbils grew well; Best rooting medium is 1/2MS+IBA0.5mg/L, the rooting rate of up to 95.6%, the average root length up to 3.1cm.3、Filaments with Lilium. sargentliae Wilson as explant in tissue culture, better filaments callus induction medium for Lilium sargentliae Wilson is MS+6-BA 0.5 mg/ L+NAA 0.5 mg/L, the callus induction rate could reach 25.6%; The best filaments adventitious Bulbils induction and proliferation of the Lilium sargentliae Wilsoncultare medium is MS + 6-BA 1.0 mg/L + 0.3 mg/L NAA, value-added coefficient reach to 6.07; Appropriate filaments adventitious Bulbils rooting medium for 1/2 ms + IBA0.5 mg/L, rooting rate up to 68.9%, the average root length 2.1 cm.4、No vaccine leaf induced by Bulbils as explant in tissue culture, blade callus induced the best medium for MS+6-BA 1.0 mg/L+1.0 mg/L NAA, induction rate can be up to 73.3%; The optimum blade adventitious Bulbils induction and proliferation of the culture medium is MS+6-BA 2.0 mg/L+NAA 0.2 mg/L, increment coefficient reach to 7.96, and adventitious Bulbils grew well; Best blade adventitious Bulbils rooting medium for 1/2 ms+IBA0.5 mg/L, rooting rate up to 97.8%, the average root length 2.7 cm.5、With 2-3cm long Lilium sargentliae Wilson tube seedlings, open it and smelting for a week. Planted into the cultivation of the soil with high-pressure sterilization after treated with 0.1% potassium permanganate, shade processing. Four kinds of explant in substrate of peat soil:vermiculite:in the cultivation of soil humus soil(1:1:1) seedlings growth state are the best, aseptic seedlings survival rate reached 100%, induced by leaf scales seedlings survival rate reached 93.3%, induced by filaments seedlings survival rate reached 73.3%, induced by bulbil induced by seedlings survival rate reached 93.3%.6、When factory production conditions is limited, bulbil relative to the filaments and scales can be used for better tissue culture explant (bulbils>scales> filaments). Bulbil can be used for large-scale factory production and germplasm preservation as explant. When a certain foundation in factory conditions, sterile leaves compared to bulbils are better explant tissue culture, sterile leaf of Lilium sargentliae Wilson as explant for large-scale factory production and germplasm preservation.
