Prunus pseudocerasus Lindl.(Cherry) is widely distributed in Hebei, Shaanxi,Gansu, Shandong, Shanxi, Jiangsu, Jiangxi, Guizhou, and Guangxi.,The leaves as themain by-products of cherry has also been used for the treatment of lessening highblood pressure.The leaves of Prunus pseudocerasus Lindl. flavonoids were extracted bymicrowave-assisted extraction technique. The effects of ethanol concentration,microwave power, microwave treatment time, microwave treatment temperature andratio of material to solvent on the extraction yields were investigated. The optimalprocess of extraction of flavones was as follows: extracting duration5min, microwavepower300W,75%ethanol,1:25(g/mL) ratio of the leaves powders to liquid, andextraction temperature80℃.The separation and purification abilities for the total flavones in the leaves ofPrunus pseudocerasus Lindl. were studied with the adsorption and desorption rates asa profile. The static adsorption and separation were used in investigation ofmacroporous adsorption resin; the dynamic adsorption and separation were used instudying the condition of adsorption and separation(the influences of the pH andvelocity of adsorption liquid, and the concentration, velocity, quantity of desorptionliquid). The availablest resin was selected and the purification effect was estimatedHPLC analysis. The results show that D101macroporous resin has the best adsorptionand desorption performance, the optimal of dynamic adsorption and separation is asfollow: adsorption liquid concentration1.0mg/mL, pH=4, The saturation adsorptionquantity is17.34mg/g (dry resins), adsorption velocity2BV/h; the reagent was70%ethanol(the three times as the volume of the resin), elutiong velocity2BV/h. thepurification rates reached74.29%,as the three times of before adsorption.Chemical compositions of the leaves of Prunus pseudocerasus Lindl. wereanalyzed and identified by HPLC-MS/MS method. Gradient elution ofmethanol-water containing0.1%formic acid was used to analyz the sample. The massspectrometer equipped with electrospay ionization source was used as defector andoperated in data was collected under the negative ion modes. Compared with thestandard sample, nine constituents were identified including seven kinds of flavonoids.With the information of MS/MS from flavonoids, the modes of breaking upflavonoids were discussed.The proposed method is rapid and reproducible and isuseful for characterization and evaluation of leaves of Prunus pseudocerasus Lindl.constituents.The antioxidant activities of the flavones in leaves of Prunus pseudocerasus Lindl. was studied regarding total antioxidant activities, superoxide anion, hydroxyl radicalscavenging activities, scavenging DPPH radical, and inhibitory ability to the peroxideof yolk lipoprotein. The results were compared with those of synthetic antioxidantssuch as ascorbic acid (Vc). The results revealed clearly demonstrated that the order ofthe hydroxyl radical scavenging activity and antioxidant activities on lipoprotein andlipid of yolk was higher than those of Vc, and the activities of total antioxidant,superoxide anion, scavenging DPPH radical were lower than Vc.
