The Mussel adhesion proteins have excellent waterproof adhesive performance and good biocompatibility. They won’t cause the body’s immune response. So they will have a wide application prospect in medicine, shipbuilding, paint and other industrigs.However, ultra-low production-and high cost through extraction approach confine their application. Now the chemical synthesis method becomes the main research in this field. At present, the most common way to introduce Dopa to molecular chain is the artificial synthesis or graft modification.In the first part of this study, a simplified route for synthesis of2-amino-3-(3,4-bis(benzyloxy)phenyl)propanoic acid hydrochloride was developed. The chemical structure of the2-amino-3-(3,4-bis(benzyloxy)phenyl) propanoic acid hydrochloride was characterized by Fourier Transform Infraredspectroscopy(FT-IR) and Hydrogen-1Nuclear Magnetic Resonance(’HNMR). Then,3-(3,4-bis(benzyloxy)benzyl) morpholine-2,5-dione was successfully prepared. The chemical structure of the3-(3,4-bis(benzyloxy)benzyl)morpholine-2,5-dione was also characterized by ’HNMR.In the second part of this study, using1-ethyl-3-(3-Dimethylaminopropyl) carbodiimide HC1(EDC) as catalyst, N-hydroxysuccinimide(NHS) as stabilizer,2-(tert-butoxycarbonylamino)-3-(3,4-dihydroxyphenyl)-propionic acid (Dopa-Boc) was grafted to silk fibroin in order to improve the adhesive performance. The chemical structure of the compound was characterized by FTIR, UV-Vis and element analysis. Adhesive performance and the stability of silk and modified silk (SF-Dopa-Boc) were investigated by the Dynamic Laser Light Scattering(DLLS), Zeta Potentials and Atomic Force Microscope (AFM). The results showed that the particle of SF-Dopa-Boc was increased in size and the absolute value of surface potential was also increased, when compared with SF in the same condition. Atomic Force Microscope (AFM) test shows that compared with SF, the contact force of SF-Dopa-Boc improve about60%.
