The degree and strength of cell adhesion and subsequent cell proliferation and differentiation depend largely on the physical and chemical properties of the surface of biological materials.The selective adhesion of artificially prepared materials to cells is usually achieved using amino acid sequences.The preparation method of the material is often complicated and not flexible.Hydrogel has attracted much attention due to its wide application in biomedical materials,cosmetics and biosensors.In this study,based on silk fibroin hydrogel materials,the interface materials of silk fibroin hydrogels functionalized with nucleic acid aptamers were constructed by covalent modification of dopamine to achieve selective cell adhesion.Using proton nuclear magnetic resonance and proton Fourier infrared spectroscopy,the successful synthesis of materials and the influence of dopamine modification on the structure of hydrogels were validated.Next,through rheological analysis and swelling rate experiments,the silk fibroin hydrogel has optimized,the strength of the gel and the method of water absorption.Further,the linker oligonucleotides were confirmed to be covalently connected to the dopamine-modified hydrogel by the click chemistry method using fluorescence imaging analysis;Finally,using the complementary sequence of the linker,nucleic acid aptamer with selective recognition of cell membrane surface proteins could be coupled to the hydrogel,and cell adhesion experiments were used to prove the selective capture and adhesion properties of the interface to target cells.Notably,the aptamer-based functionalization strategy can be extended to other cell-surface receptors by coupling one or multivalent aptamers through different customized Linker sequences.The versatile functionalization could be used to regulate the dimerization and oligomerization of the receptors,affecting cellular behaviors.Together,this article proposes a new nucleic acid aptamer functionalized hydrogel construction strategy,which can be used as a general platform to enhance the interaction between biomaterials and cells(such as selective adhesion,cell migration or polarity).Nucleic acid aptamers can efficiently regulate cell behavior at the level of aptamers.This strategy is expected to be applied in tissue engineering and regenerative medicine.
