By taking capsaicin as the research object, the conditions of the capsaicin extraction by decompressing innner ebullition were optimized by using response surface, and the influence of these conditions on extraction yields of capsaicin were discussed. The preparation technology of capsaicin-chitosa/Sodium Alginate Microsphere (CCSAM) was timized by means of the single-factor experiment, and the release effects of CCSAM were researched. This study not only provided technology references for the extraction and purification of capsaicin, but also had good industrial prospects.The results of response surface optimization experiments were as follows. The concentration and the amount of desorption solvent, and the concentration of extraction solvent had significant influence on the extraction yields of capsaicin, and the important order of above conditions was, and the concentration of extraction solvent> the amount of desorption solvent> the concentration of desorption solvent. Besides the amount of extraction solvent and the extraction temperature had no significant influence on the extraction yields of capsaicin. The optimum technological conditions of CDIEET were that, the concentration and the amount of desorption solvent were82%and3.5mL·g-1respectively, the concentration and the amoun of extraction solvent were44%and12mL·g-1respectively, the extraction temperature was50℃. And the optimum yield of capsaicin was0.212%which was approximately equal to the capsaicin yieid of verification test. Compared with extraction by ethanol refluxing extraction, CDIEET has following advantages, higher yield of capsaicin, better effect of subsequent purification, less destruction of material and so on.The results of the preparation of CCSAM were as follows. Under the optimum conditions that, the concentration of sodium alginate was15.00g·L-the amount of calcium chloride solution was4times volume, and the amount of chitosan solution was2times volume, the CCSAM was prepared successfully, of which the entrapment efficiency and drug loading was43.0%and18.2mg·g-respectively. The study of the vitro release mechanism of CCSAM showed that, capsaicin incorporated CCSAM was not released in the artificial gastric juice, and released according to the first-order releasing kinetics in artificial intestinal, it has good slow release efficiency.
