| Lotus Plumule is the young leaves and seeds radicle in mature seeds of plantLotus which belong to water lily family of the genus (Nelumbo nucifera Gaertn). It isa important traditional Chinese herbal medicine, and has pharmacological researchvalue. JianNing, a city in Fujian is “the hometown of Jian Lotus†and is one of themain producing area of Lotus Plumule. Extraction technology, purificationtechnology, the anti-oxidantive activity and the antibacterial activity of totalflavonoids from Lotus Plumule were studied in this paper. Therefore, an importantrecommedation were offered for developing deep-processing products of totalflavonoids from Lotus Plumule.(1)An solvent extraction process of total flavonoids from Lotus Plumule wasstudied in this experiment. Different factors were considered, such as liquid-solidratio, the solvent, extract time, pH value, extract temperature. And basing on thesingle factor tests, the orthogonal tests were done. It indicated that the optimumparameters about cellulase extraction were that ethanol concentration is75%,liquid-solid ratio is50:1, extract temperature is75℃and extract time is2.5h, then theextraction rate could reach to4.20%.(2)The extraction process of total flavonoids from Lotus Plumule by enzymatichydrolysis of cellulase was studied in this experiment. Different factors wereconsidered, such as zymolysing reaction time, temperature, pH value and so on. Andon the basis of single-factor experiments, the optimum parameters were set up by theresponse surface. The optimum parameters about cellulase extraction were concludedas follows: ethanol concentration50%, liquid/solid ratio19:1, enzyme adding amount0.3%(m/m), pH5.5, reaction temperature60℃, reaction time2h, the actual detectionvalue of flavonoids extraction rate was4.50%. The extraction rate of enzymatichydrolysis of cellulase was higher7.14%than solvent extraction.(3)The characteristics of macroporous adsorption resins S-8, X-5, AB-8, D101,and HP-20on static adsorption and desorption rate of total flavonids from LotusPlumule were investigated. And the result showed that X-5resin has higher absorption and desorption rate. The optimization of purification conditions of X-5resin were concluded as follows: sample concentration was about5.1mg/mL, pH was6, the sample flow rate was2BV/h, the sample quantily was2BV, eluent was50%(V/V) ethanol solution, eluent flow rate was2BV/h, eluent quantily was4BV. Thenthe purity of total flavonoids was50%. And it could be a suitable alternative toindustrial production because of its easy handling, effective and low cost.(4)The total antioxidant capacity and the effect on scavenging·OH, DPPHand·O2-radicals of total flavonoids from Lotus Plumule were studied, which werecompared with the effect of Vc. The result showed that the total flavonoids fromLotus Plumule could effectively scavenge·OH, DPPH and·O2-, and have the strongtotal antioxidant capacity. Meanwhile, the total flavonoids from Lotus Plumule hasstronger scavenging ability on·O2-and higher total antioxidant capacity than Vc. Andcompared with Vc, it has lower ability on·OH and DPPH radicals.(5)The antibacterial activity of total flavonoids from Lotus Plumule wasstudied, which was compared with the effect of Rutin. It showed that the totalflavonoids from Lotus Plumule had no antibacterial activity against Penicillium andRhizopus. But it had a significantly positive inhibitory activity against Staphylococcusaureus, salmonella, Escherichia coli, and Bacillus subtilis. Meanwhile, the totalflavonoids from Lotus Plumule didn’t be affected by changing temperature andchanging pH value. In addition, the MIC for Staphylococcus aureus is1mg/mL; theMIC for salmonella is0.5mg/mL; the MIC for Pseudomonas is2mg/mL; the MIC forEscherichia coli is4mg/mL; the MIC for Bacillus subtilis is4mg/mL. |
