It is a kind of natural medicines and health products with along history in our country. It was used for cleaning heat, reducing phlegm, stoppingblood and anti-inflammatory. In research, the key health-care components in the seedsof Nelumbo Nucifera Gaertn can better debase blood pressure, antagonize arrhythmia,etc. They have wide therapy foreground in desease of blood vessel in heart and brain.Therefore, as a kind of natural resources of broad-spectrum, highly effective, non-toxicside effects, the importance of its extraction, separation and purification was consideredby increasingly science and technology workers. This paper adopts a new method,surfactant and ultrasonic-assisted, to extract total alkaloids from lotus plumule forimproving the extraction rate. Moreover, clarify the extract with chitosan, and discusssome factors that affect the stability of alkaloids. Thesis mainly includes the followingaspects:Established a method for determination of the content of liensinine-Uvspectrophotometry. Through to absorption curve, the wavelength275nm for liensininedetection is determined. And the concentration from20.0to80.0μg.mL-1has a goodlinear relationship for absorbance value, the regression equation as y=0.0152x-0.0025,R2=0.9998. Then through to determin the precision test, the stability test, therepeatability test, the recovery test, within-day precision test, day to day precision test.The experimental results show that it can maintain stable within24h, within-dayprecision and day to day precision is good, reproducibility and recovery rate areaccordance with the requirements of the methodology.Optimize the process of surfactant and ultrasonic-assisted extract alkaloids fromlotus plumule. The paper discussed the different factors effect on the extract rate bydifferent material condition, solvent, surfactant category, surfactant concentration,solid-liquid ratio, ethanol concentration, ultrasonic power, ultrasonic time, soak timebefore extraction, extracting times. On the basis of single factor experiment, using theuniform design and orthogonal design to optimize extraction process. The paperidentified the best extraction conditions:2%APG0810, solid-liquid ratio1:50, ethanolconcentration70%, ultrasonic power105W, ultrasonic time25min, soak time before extraction20min, ultrasonic extraction time1. Under these conditions, the extractionrate is140.024mg/g. Compared to heat reflux extraction (96.341mg/g), soxhletextraction (115.762mg/g), and simple ultrasonic extraction (122.793mg/g), the processof surfactant and ultrasonic-assisted extraction has a certain advantage.Optimize the process of extract clarification with chitosan. The paper studieddifferent factors effect on removing impurity by the dosage of chitosan, stirringtemperature, concentration of the extract, mixing time, and pH, using single factormethod coupled with orthogonal design. Results showed that the optimum conditionswere: chitosan dosage1.2mL/g, stirring temperature40℃, the concentration of theextract1:5g/mL, stirring time20min, pH6.27process conditions, under which theprocess of purifying is the best and stable.Study the factors that affecting the stability of alkaloids. In this section, pH, ionicconcentration, light and temperature were considered. The results indicated that,compared to in light and solution state, the stability of alkaloids in dark, roomtemperature and solid state is better. If saved in solution state, we′d better ensure theenvironment of solution keep low ionic concentration and pH5. |
