| In this paper, yeast species and genetic diversity in folk sourdough and its relatedsubstrates were investigated. Totally,1040yeast strains were obtained from folksourdough and its related substrates using alcohol contained enrichment medium,diluting and spreading on plates directly or after fermentation. The isolated strains werefirst classified morphologically. Then the ITS1and ITS1p regions of strains wereanalyzed by single-strand conformation polymorphism (SSCP). And the representativestrains of distinct bands were further identified by sequencing the ITS and D1/D2regions. After blasting on NCBI, all the isolated strains were attributted to11genera and30species including283S. cerevisiae strains. Among them, one new Kazachstaniaspecies were analyzed in detail by means of conventional methods and moleculartaxonomy.The hypervariable intergenic spacer1(IGS1) region of ribosomal DNA of S.cerevisiaes strains from sourdough were studied by SSCP analysis. Consequently, ahigh genetic diversity of S. cerevisiaes strains from folk sourdough was found. Also byMLST analysis of S. cerevisiae strains from different sources, phylogenetic tree wasconstructed and the evolutional relationship among populations of folk sourdough andother sources was recovered. The results indicated that S. cerevisiae strains from folksourdough belonged to an independent domesticated clade, which differentiatedapparently from primeval forest strains.This study enlarged the number and sources of S. cerevisiae strains from folk sourdough and related substrates, which shed light on the origin of S. cerevisiae fromfolk sourdough and also accumulated a great deal of strain resources for their furtherexploitation and utilization. |
