| Wine yeast convert sugars into ethanol,carbon dioxide and hundreds of other secondary metabolites,and they also affect wine flavor.While most yeast strains during winemaking are classified as Saccharomyces cerevisiae.Different S.cerevisiae strains are suitable for the different fermentation process so studying the genetic diversity of S.cerevisiae strains is of great significance for wine production and selection of S.cerevisiae strains with good characters.192 S.cerevisiae strains isolated from viticultural areas of Xinjiang and Ningxia were typed by four microsatellite markers,and the genetic relationships among these strains were also analyzed.The results were as follows:1.The results showed the diversity of S.cerevisiae strains in Xinjiang regions.A total of 54 alleles,which generated 29 genotypes,were found among 47 strains from Xinjiang region;the mean allelic number of per locus was 13.5,the most was 15(SCAAT1)and the least was 12(SCYOR267c);the polymorphism information contents(PIC)of all strains at four loci were 0.7869~0.8479,which>0.5000,suggesting that the four SSRs(simple sequence repeats,SSRs)loci were hypervariable;the heterozygosity expected and heterozygosity observed were 0.8193~0.8714 and 0.8511~0.9787,respectively.2.The results showed the diversity of S.cerevisiae strains in Ningxia regions.A total of 160 alleles,which generated 123 genotypes,were found among 145 strains from Ningxia region;the mean allelic number of per locus was 40,the most was 47(SCAAT1)and the least was 29(YPL009c);the polymorphism information contents(PIC)of all strains at four loci were 0.8939~0.9453,which>0.5000,suggesting that the four SSRs loci were hypervariable;the heterozygosity expected and heterozygosity observed were 0.9046~0.9509 and 0.8483~0.9793,respectively.3.The Fst values(Fixation indices of individuals relative to the total population)of two groups were 0.0355~0.0423,which suggests more obvious genetic differences between strains in each population than between two populations.4.A total of 39 alleles,which generated 11 genotypes,were found among 16 strains with SSRs markers while a total of 17 alleles which generated 5 genotypes were found among 16 strains with Interdelta sequences. |
