Plant suspension cultured cells, a proven physiological and versatile cell system, are widely used in high-throughput analysis and functional characterization of genes. Puccinellia tenuiflora, is not only a special kind of salt tolerance of wild herbs, and also a potential resistance gene banks。Some heterologous systems have been used for characterization of Puccinellia tenuiflora genes, a complete homologous expression systems have not been reported。Here, we report a simplified method for establishment of Puccinellia tenuiflora suspension cultured cells, and optimization of culture conditions. We get the well dispersed and rapidly growing suspension culture cell-free system., and as the material to established a highly efficient and rapidly way of stable transgenic system. Not only that, This system is applied to the study of the subcellular localization,and obtained excellent results. This transformation system is not only homologous system for characterization of Puccinellia tenuiflora genes, but also be used for the parsing of the functional genes of other species. Due to the transformation cycle is short, There are variety of purposes in addition to research for the subcellular localization. And can be used as a research model of the wild plants or renewable difficult plant.We show here a simplified and highly efficient stable gene expression system using Puccinellia tenuiflora suspension cultured cells and its applications in protein subcellular localization assays. These transgenic suspension cultured cells will be particularly useful in studies of protein location and shape in cells.
