Puccinellia tenuiflora, one of the halophytes, is considered as ideal plant material for the study of salt tolerance mechanism and cloning of salt tolerant genes. PtMAPKK6 is an important member of MAPK signaling pathway in Puccinellia tenuiflora. In the previous works of our lab, the EST fragment of PtMAPKK6 gene was isolated by suppression subtractive hybridization and the cDNA sequence of PtMAPKK6 was cloned by RACE technology. In this study, the open reading frame sequence of PtMAPKK6 gene was cloned by RT-PCR from Puccinellia tenuiflora; expression characteristics of PtMAPKK6 gene in Puccinellia tenuiflora roots in response to salt, drought, and low temperature stress were analyzed by qRT-PCR, the expression patterns of PtMAPKK6 and other salt responsive genes including PtCytatin, PtMatK, PtHATPG, PtLIR1 and PtSS were compared. The results indicated that PtMAPKK6 extensively participates in the response of Puccinellia tenuiflora roots to salt, drought, and low temperature stress, and may play important roles in the early response to adverse environment stress.
