Nowadays, the diseases of blood coagulation system have been concerned for their mortality and morbidity. There are more than five million patients who were suffering for this kind of diseases every year in China. Many a pharmaceuticals are required to be researched and developed. Compared with congener products, venom, as a kind of biological preparation, has lots of advantages such as high efficiency, fewer by-effects, and secure usage, and would be extensive applied in clinic. However, the researches about the utilization of venom enzymes are most focused on a single enzyme. It would waste many biological resources. Therefore it is necessary that as many as possible enzymes should be isolated from the same venom preparation.Agkistrodon halys ussuriensis venom was fractionated by chromatographies with ion exchange chromatogram,Sephacryl S-100 chromatogram and affinity chromatogram respectively. The fibrinolytic enzyme was subjected to SDS-PAGE. The result of the experiment shows a single band on SDS-PAGE. The molecular mass of fibrinolytic enzyme is 31.4kD, and its isoelectric point is 6.4 by using equipotential electrophoresis. The result shows that the protein recovery of fibrinolytic enzyme arrives to 2.5%. It had both fibrinolytic enzyme activities and arginine esterase enzyme activities.This article describes a simple method to isolate fibrinolytic enzyme from the same venom preparation after the purification of thrombin-like enzyme. The method we describe here is an efficient approach for the comprehensive utilization of the venom.
