Objective: Explore the relationship between serum CK-MB, pathological changes of myocardial, expression of TNF-αon myocardium tissue of mice and different dose of venom, the poisoning time induced by Agkistrodon Halys Pallas venom.Methods: (1)Using two-factor factorial design, the 96 male Kunming mice of clean grade were divided into 12 groups(n=8) randomly. Snake venom was divided into three levels: 0-dose(0.0mg/kg), low dose (0.5mg/lg), high-dose (1.0mg/kg); Poisoning time to set up four levels : 3 hours, 6 hours, 24 hours, 48 hours.(2)Collected serum and myocardial specimens, detected of CK-MB by biochemical analyzer, investigated pathological changes of myocardial, detected TNF-αprotein expression of myocardial tissue by Immunohistochemical.(3) Statistical Methods: experimental data using the SPSS 13.0 statistical software for statistical analysis, measurement data were presented as ( x±S) , data analysis using factorial, correlation analysis using linear correlation analysis, P<0.05 regarded as statistically significant.Results: (1) The test group serum CK-MB value was significantly higher than the control group were, the difference was significant (P<0.05), high-dose group was higher than the low-dose group, the difference was significant (P<0.05). (2) Through various periods of the statistical analysis of myocardial histopathologic scores, high-dose group than in the low dose group of serious diseases, the difference was statistically significant (P<0.05), 48h group than in the 3h, 6h group of serious diseases, the difference was statistically significant (P<0.05),as the dose increased, the time to extend ,the higher points of myocardial lesions, proved that snake venom can lead to myocardial cell degeneration and necrosis, and as the venom dose increases, the more severe myocardial injury, it had time and dose-response relationship. (3) Monitored by immunohistochemistry in mice with myocardial expression of TNF-αprotein was found that compared with the control group, experimental group of mice myocardial TNF-αprotein expression was significantly increased, the difference was statistically significant (P<0.05). (4)correlation analysis: CK-MB value and myocardial histopathologic scores correlation analysis, showed a positive correlation (r=0.966,P<0.05);TNF-αprotein expression and myocardial histopathologic scores correlation analysis, showed a positive correlation(r=0.869, P<0.05).Conclusions:(1)Agkistrodon snake venom can lead to acute myocardial injury of mice and elevated CK-MB, the more severe myocardial injury, the more CK-MB values increased significantly. (2)with acute myocardial injury of mice induced by Agkistrodon Halys Pallas venom,expression of TNF-αon myocardium tissue of mice was increased,and the increase of the dose of venom and the poisoning time,the more expression of TNF-α,showing a certain time,dose effect trend,suggesting that overexpression of TNF-protein one of the mechanisms of acute myocardial injury induced by Agkistrodon Halys Pallas venom.
