Purpose: Looking for the active substance of whitening effect in ginseng, Extraction andseparation and purification of the effective ingredients by modern biotechnology means.Method: Determination of whitening substance on tyrosinase activity of the inhibitionrate, application biochemical enzymology methods, and optimization the best experimentalcondition. Using different extraction methods, different ratio of solution, different extractiontime, and different extraction times to extract the ginseng whitening crude extracts, to thetyrosinase activity of the inhibition rate for the evaluation standards, and optimization the bestextraction methods. Application of alcohol precipitation method, silica gel columnchromatography, Sephadex LH-20column chromatography, C18reverse phase silica gelcolumn chromatography, high performance liquid chromatography and other technologies,separation and purification on the whitening active substances of ginseng.Result:1Optimize the determination of tyrosinase activity of the inhibition rate to the bestconditions for the experiments were as follows: the concentration of L-tyrosine was0.0754mg/mL, the concentration of tyrosinase was0.1082mg/mL, constant temperature time was10minutes and reacting time was5minutes;2Optimize of the extraction of ginseng whitening crude extracts to the best extractionmethod were as follows: using water decocting methods to extract for3times, each extraction2hours, liquid ratio of1:10, using this method to extract the ginseng whitening crude extractson tyrosinase activity of the inhibition rate was8.11%, the yield was9.73%;3Using different alcohol precipitation method to separation ginseng whitening crudeextracts, optimize of the step-by-step gradient method is the best alcohol precipitation method,separation get the ginseng of the ethanol soluble materials on tyrosinase activity of theinhibition rate was18.61%, the yield was3.38%;4Ginseng of the ethanol soluble materials separated by silica gel column chromatographyand get No. Ⅱ andⅢ effective ingredients, to the tyrosinase activity of the inhibition rate ofrespectively37.99%and37.32%, No.Ⅱ and Ⅲ of the merger components yield of0.27%;5The effective components by Sephadex LH-20column chromatography get D ingredients, to the tyrosinase activity of the inhibition rate of76.79%, the yield was0.058%, through tothe D ingredients in the UV wavelength scanning, found the maximum absorption at the278nm;6The D ingredients by C18reverse phase silica gel column chromatography get effectiveingredients D6, to the tyrosinase activity of the inhibition rate of83.25%, the yield was0.012%, after the purification of the effective ingredients by thin layer chromatography test get oneof spot, HPLC detection for a single spectral peak, the retention time of9.32min.Conclusion: Application biochemical enzymology methods tracking detection theeffective ingredients on the tyrosinase activity of the inhibition rate, the establishment of astable, viable extraction and separation and purification process for ginseng whitening activesubstances. After the purification of the active ingredient were reached the chromatographicpure, and have good whitening effect. The study provides scientific basis for ginseng in theapplication of whitening cosmetics, at the same time for the further development of theginseng whitening cosmetics which laid the experimental basis.
