Investigation On Whitening Effect Of Yupingfeng Powder And Preparation Of Its Gel

Yupingfeng Powder is a famous tonifying prescription,which has the function of benefiting qi and consolidating the surface and stopping sweat.The theory of traditional Chinese medicine believes that the body surface is deficient in qi,and the face cannot be nourished by qi and blood.It will appear dark spots and yellow.Therefore,in this paper,based on the whitening theory of traditional Chinese medicine,Yupingfeng Powder was taken as the main research object,and the whitening efficacy was taken as the index.The optimal drug ratio of whitening efficacy was screened by uniform design method,and then the extraction process was optimized.At the same time,the mechanism of Yupingfeng Powder on whitening efficacy was further studied,and finally the whitening gel product was prepared.The main research contents and results are as follows:（1）Taking tyrosinase inhibition rate,DPPH free radical clearance rate and ABTS+clearance rate as whitening evaluation indexes,the best drug ratio of whitening effect of Yupingfeng powder was investigated by using uniform design method.The results showed that the best drug ratio of Yupingfeng powder was Fangfeng:Atractylodes macrocephala:Astragalus membranaceus was 33:95:72,the inhibition rate of tyrosinase activity was 46.45%,the clearance rate of DPPH free radical was 90.22%,and the clearance rate of ABTS+was 90.52%.（2）Taking the whitening effect as the index,the material liquid ratio,extraction solvent and extraction temperature were selected as the investigation factors to study the best extraction process.The experiment of three factor and three-level response surface analysis method was established,and the optimal process of Whitening Extract of Yupingfeng powder was optimized through the comprehensive analysis method of multiple regression model:solidliquid ratio 1:9g/mL,ethanol concentration 65%,extraction temperature 92℃.（3）In the study on the whitening mechanism of the extract of Yupingfeng powder,it was found that the extract of Yupingfeng powder had no inhibitory effect on tyrosinase monophenolase,only diphenolase,and the half inhibitory concentration IC50 was 342.65mg/mL.Further analysis of tyrosinase inhibition showed that the inhibition mechanism was reversible inhibition,and the extract combined with free enzyme and enzyme substrate complex,which belonged to mixed inhibition type.The inhibition constants Ki and kis of free enzyme and enzyme substrate complex were 0.0034mg/mL and 0.0071 mg/mL.（4）In the preparation of Yupingfeng whitening gel,sensory evaluation and physical and chemical properties determination were used as evaluation indexes to screen the selection of gel matrix,the addition amount of moisturizer and the addition amount of Yupingfeng whitening extract.Finally,it was concluded that the gel matrix was composed of carbomer-940 and hydroxypropyl methyl cellulose in the same proportion.The addition amount of moisturizer was 1%butanediol,1%hyaluronic acid,2%glycerol,and 0.05%EDTA-2Na;the optimal amount of Yupingfeng whitening extract was 2%.（5）Through safety evaluation,Yuping wind whitening gel has no skin irritation and no skin toxicity.Finally,the effect of Yuping wind whitening gel was tested.It was concluded that the best ratio of Yuping wind whitening gel 6 weeks later,the melanin value decreased by 16,with excellent whitening effect.
