Objective To investigate the expression and dynamic changes of Wnt3 a,?-Catenin,OPG,RANKL under different degrees of inflammation in chronic periapical periodontitis.Methods A total of 30 normal and chronic apical periodontitis specimens were selected and divided into control group,mild to moderate inflammation group,and severe inflammation group using hematoxylin-eosin(HE)staining,with 10 samples in each group.Immunohistochemistry was used to detect the dynamic expression of Wnt3 a,?-Catenin,OPG,and RANKL under different levels of inflammation in chronic apical periodontitis,and to observe its dynamic expression in neutrophils and lymphocytes.Results1.Wnt3 a,?-Catenin,OPG,RANKL were positively expressed in chronic periapical periodontitis.Compared with the control group,except that the expression level of Wnt3 a in the chronic periapical periodontitis group was reduced and there was no statistical difference(P> 0.05),?-Catenin,OPG,RANKL were all increased in chronic periapical periodontitis,with statistical differences(P <0.05).2.According to the degree of HE staining of inflammatory cell infiltration,the components of chronic periapical periodontitis were divided into mild and moderate inflammation group and severe inflammation group for immunohistochemical staining.The results showed that:compared with the control group,the expression level of Wnt3 a varies with the degree of inflammation Increased and showed a downward trend and then increased(P<0.05);?-Catenin,OPG,RANKL in the control group,mild to moderate inflammation group,severe inflammation group showed different degrees of increase with the increase in inflammation(P<0.05);The ratio of RANKL/OPG changes first and then decreases as the severity of inflammation increases.3.In the periapical periodontitis group,the expression levels of Wnt3 a,?-Catenin,OPG,RANKL increased with the severity of inflammation,and the difference was statistically significant(P <0.05).In the apical periodontitis group,the expression level of Wnt3 a increased with the severity of inflammation,and the difference was not statistically significant(P>0.05).The expression levels of ?-Catenin,OPG and RANKL also increased with the severity of inflammation,and there were statistical differences(P <0.05).Conclusion Wnt3 a,?-Catenin,OPG and RANKL were dynamically expressed in different degrees of chronic periapiculitis,and the ratio of RANKL/OPG changed.Wnt3 a,?-Catenin,OPG,RANKL may play a direct or indirect role in the course of chronic periapical inflammation.
