| ObjectiveNasopharyngeal Carcinoma is one of the head and neck cancer in China, and has strong genetic predisposition. This study chosen cyclin-dependent kinase inhibitor CDKN2A gene and BARTs which widely expressed in Epstein-Barr virus infection, to investigate the relationship between single Nucleotide Polymorphisms of CDKN2A,EBV A73gene and nasopharyneal carcinoma in a Yunnan population. At the same time find out the contact with Snps and clinical parameters.MethodsCDKN2A gene variant rs1412829was genotyped in195individuals consisting of95NPC cases and100normal controls (NC) using TaqMan genotyping allelic discrimination assay. EBV A73gene variant A157154C was genotyped in103individuals consisting of51NPC cases and52normal controls using nested polymerase chain reaction genotyping allelic discrimination assay.Results1. The frequency of the G allele was significantly higher in the NPC group than that of control group (20%vs.9%, P=0.023). And compared with A subjects, G carriers of rsl412829had a higher risk of NPC (OR=2.497[1.107~5.632], P=0.031).2. The genotype frequency of the G allele was also remarkably higher in the NPC group (P=0.031).3. The frequency of the G allele was significantly higher in the advanced stage group than that of early stage group (P<0.001).4. The frequency of the C allele was significantly higher in the NPC group than that of control group (96.1%vs.51.9%, P<0.001). The genotype frequency of the C allele was also remarkably higher in the NPC group than that of NC group (96.1%vs.42.3%,P<0.001).5. Compared with AA subjects, CC carriers of A157154C had a higher risk of NPC (OR=22.685[7.772-66.217],P<0.001).6. The frequency of the C allele was significantly higher in the male group than that of female group (73.2%vs.26.8%,P<0.001).ConclusionsCDKN2A gene rs1412829SNP is associated with NPC and GG genotype of CDKN2A gene polymorphism modulates nasopharyngeal carcinoma. Besides, this SNP is related to pathological stage, we may consider that G allele was found easily in the advanced stage of nasopharyneal. EBV A73gene A157154C SNP is associated with NPC and CC genotype of EBV A73gene polymorphism modulates nasopharyngeal carcinoma. In addition, this SNP is related to gender, it might be one of the reasons for that men had a significantly risk of NPC than women do. |
