Objective:Taking Traditional Mongolian Medicinal herb Rubus sachalinensis Leveille as research object, we analysis its viral activity, anti-bacterial activity and anti-inflammatory biological activity. After extracting and purifying the extract from ethyl acetate and n-butanol layer with the function of anti-bacterial and anti-inflammatory, we discover new compounds and active monomer compounds with the method of1H-NMR,13C-NMR,MS and structure identification. The research provides a scientific base for the research of pharmacodynamic and quality control.Methods:Air-dried aerial part of Rubus sachalinensis.(5.05kg) was chipped and refluxed in75%Ethanol three times and refluxed in95%ethanolone one time. The ethanolic extract was decanted and combined; the solvent was evaporated at reduced pressure and obtain extract0.924Kg.It will be used to extract the same amount of water, petroleum ether, chloroform, ethylacetate, n-butanol in turn extraction of three separate the solvent was evaporated at reduced press. After the remnant water on polyamide column chromatography concentrated, ordinal water, methanol-water (1-1), methanol elution and decompression recovered solvents. Every parts were measured by the method of brine shrimp lethality to track bioactivity, and the select the most active part to do next research. Using Silicagel colureversed phase preparative HPLC, Sephadex LH-20to separate and purify integrates and to get compounds in pure form. Using spectroscopic methods including1H-NMR,13C-NMR, HMBC, MS to establish the structures of the isolated pure compounds.Result:In this thesis,31compounds were obtained from the EtOAC and n-butanol. Their structures were elucidated by spectroscopic methods including1H-NMR,13C-NMR, MS and chemical methods. As a result, there are12triterpenoids(1-12),8flavonoids (20-27),4lignans (28-31)and7(13-19)other compounds.These compounds were identified respectively as follows:2α,3β,19α-trihydroxy-olean-12-en-24-CHO-28-CO2-glucopyranoside (1),2α,3β,19α-trihydroxy-urs-12-en-24-CHO-28-CO2-glucopyranoside (2),2α,3β,19α-trihydroxy-urs-12-en-24-CHO-28-CO2- glucopyranoside (3),2α,3β,19a-trihydroxy-urs-12-en-24-COOH-28-CO2-glucopyranoside (4),2α,3β,19a-trihydroxy-urs-12-en-23-CHO-28-CO2-glucopyranoside (5),2α,3β,19a,24-tetrahydroxy-urs-12-en-28-CO2-glucopyranoside (6),2α,3β,19a-trihydroxy-urs-12-en-28-CO2-glucopyranoside (7),2α,3β,19a-trihydroxy-urs-12-en-28-oic acid (8),2α,3β,19α-trihydroxy-urs-12-en-24-CHO-28-oic acid (9),Euscaphic acid (10),2α,19α-dihydroxy-2-oxo-urs-12-en-28-oic acid(11),3α,19α-dihydroxy-urs-12-en-28-oic acid(12), Gallic acid (13),Caffeic acid (14),Ethyl gallate (17),Quercetin (20),Kaempferol (21),quercetin-3-O-β-D-glucopyranose (22),quercetin-3-O-α-L-glucopyranose (23*),Tiliroside(24),Cis-tiliroside(25),1,2,3-gallayl-α-D-glucopyranose (30),1,2,3-gallayl-β-D-glucopyranose (31)。Conclusion:This research on Traditional Mongolian Medicinal herb R. sachalinensis's effective active part of ethyl acetate and n-butanol extraction layer. The chemical were separated and31compounds were gain. Among them,4(23,26,28,29) compounds are new compounds and13(1,2,3,4,6,7,9,10,11,12,27,30,31) compounds were isolated from this plant for the first time. Therefore, this research not only helping identify the bioactive components of R. sachalinensis, but also giving an instruction of medicine-making with R. sachalinensis. The research expands natural product libraries and also explains effectiveness of R. sachalinensis in field of modern biomedical.
