| The genus Gentiana (Gentianaceae) consists of about400species distributedthroughout the world, especially in mountain areas of the north temperate zone andtropical zone. There are about22genus,500species grown in China, and12genus,70species are used in medicine. According to Chinese Pharmacopoeia, original plants ofGentiana are Gentiana scabra Bunge, Gentiana triflora pall., Gentiana manshuricaKitag. and Gentiana rigescens Franch.. Gentiana is a commonly used traditionalChinese medicine. It has the functions of protecting liver, cholagogue, invigoratingstomach, exciting central nerve, anti-inflammatory and antibiosis.The thesis is a systematic investigation on the chemical constituents andbiological activity of Gentiana farreri Balf. f. First of all, Gentiana farreri wassoaked in ethanol, we got the ethanol extract. Then the ethanol extract was extractedwith petroleum ether, acetic ether and n-butanol in order and we got extract ofpetroleum ether, extract of acetic ether, extract of n-butanol. The extracts ofpetroleum ether and acetic ether were subjected to silica gel, column chromatography,MCI, sephadex LH-20and polyamide column chromatography, finally28compoundswere isolated and purified. The structures of22compounds were elucidated by EI-MS,IR,1H-NMR,13C-NMR, DEPT spectroscopies. These compouds belong to triterpene,flavone, steroid and so on.The22compounds isolated from Gentiana farreri were identified as linoleic acid(1), β-sitosterol (2), pseudotaraxasterol(3), α-amyrin(4), β-amyrin(5),28-hydroxyl-α-amyrin(6),28-hydroxyl-β-amyrin(7),3β-palmitate-28-hydroxyl-α-amyrin(8),3β-palmitate-28-hydroxyl-β-amyrin(9), lupeol(10),28-hydroxylupeol(11),24-hydroxy-lupeol(12), β-daucosterol(13), palmitic acid(14), ursolic acid(15),2α,3-dihydroxylu-rsolic acid(16),2α,3-dihydroxyloleanolic acid(17), dibutylphthalate(18), p-hydroxybenzaldehyde(19), hydroxyacetophenone(20), isoscoparin(21), isoorientin(22).Allthese compounds were isolated from G. farreri for the first time.In addition, the antimicrobial activity and anti-tumor activity of all theextracts and partial compounds were determined by Oxford cup and MTT assay forthe first time. The results show that the extracts of acetic ether and n-butanol havefine antimicrobial activity, but the extract of petroleum ether only show antimicrobialactivity to partial bacterium. Meanwhile, all the extracts and compounds show variedanti-tumor activity to HepG-2cells, compound9,16,17,18have high activity, IC50is 67.52,13.03,13.19,41.26(μg/mL), respectively.The thesis reviews the chemical constitutents and pharmacological activity of thegenus Gentiana and conducts research on chemical constitutents and bioactivity ofGentiana farreri Balf. f.. We hope this work could provide the relevant scientific basisfor the development and utilization of natural medicine. |
