Qinjiao is roots of large leaved gentiana (Gentiana macrophylla Pall.) which belongs to Genus Gentiana, Family Gentianaceae. It is genuine crude medicine of Shaanxi Province, and also an important traditional Chinese drug with a long history of use for dispelling the wind, relieving rheumatic, eliminating pyretic, stopping numb and pains.To ulteriorly control the quality, and establish the fundamental for clinical use and new drug development of Qinjiao as well, we carried out a systematic research on the chemical constituents of Qinjiao collected from Long county Shaanxi province. Eleven compounds were achieved using several separation methods. Nine of them were elucidated by the means of spectroscopy(UV, IR, MS, 13C-NMR, 1H-NMR, HMBC, HMQC, HH COSY etc.) assisted by comparing with standard substance and constants. Five oil dissolvable constituents were: ( A1 ) erythrocentaurin, (A2) roburic acid, (E) oleanolic acid and two novel compounds (B) and (D) . (B) was named as N-pentacosyl-2-carboxy-benzoyl amide and (D) was named as erythrocentaurin acid. Water dissolvable fraction gave four known secoiridoid glucosides. They were: ( S1 ) gentiopicroside, ( S2 ) swertiamarine, (S3) sweroside and (S4) 6'-O-β-D-glucosylgentiopicroside. Among the known compounds, (A1) erythrocentaurin was isolated from genus Gentiana for the first time.Furthermore we did a series work on investigating the content of gentiopicroside, the mark constituent in Qinjiao and some rule about accumulating and variation of it. Though the studies on fingerprinting chromatography by HPLC method, we found the character of this crude medicine from Long county and a common template was set up.
