In order to understand the chemical constituents and bioactivity of second metabolites of Rhizobium, ethyl acetate extraction of fermentation of Rhizobium CCNWGX022 and distillate fermentation fluid were studied.Through separating the ethyl acetate extraction by silica gel column, three compounds were gained and named IV, VII, and VIII. IV and VII were determined asα-amino acid-unsaturated lactone and terpenoid respectively using color-changing reaction. The structure of compound VIII was identified by spectral evidence, and elucidated as hexadecanoic acid on the basis of EI-MS, 1HNMR, 13CNMR, DEPT and H-HCOSY spectral evidence.The analysis of chemical constituents showed the existence of steroids, terpenoids, amino acids, aminosugers, phenols, alkaloids and volatile oils in distillate fermentation fluid. GC-MS analysis indicated 27 compounds in distillate fermentation fluid. Most of them belonged to alkyl, fatty acid esters, phthalate esters, and phenols.The distillate fermentation fluid did not show significant antibacterial or anti-epiphyte activity. Through the tests it only affected the growth rate of Colletotrichum gloeosporioides as a promoter at first 2 days but an inhibitor at the rest of time. The distillate fermentation fluid affected the germination and root growth of lettuce, a non-nodulateable plant of CCNWGX022. It delayed germination process on seeds, inhibited long-growth of root, but promoted development of root hair, inhibited its desquamation, enhanced the activity of root significantly.
