Objective:The aim of this research is to study the pathogenesis of COPD and find out a effective treatment,. according to the current research of relationship between apoptosis and COPD, we research the expression of apoptosis genes in rats with COPD, while treating rats of chinese Nationality's herbs and observing the expressive changes of apoptosis genes and inhabitor of apoptosis after the treatment.Methods:1.The COPD rat model was established by exposure to cigarette smoking and inhalation of lipopolysaccharide twice a month.2. We mesure the pathological changes of lung tissues by semi-quantitative pathological analysis.3. Immunohistochemistry and RT-PCR were used to measure the apoptosis genes (caspase-3/survivin) in protein and transcription level and the difference of expression between COPD group and miao nationality's herbs group were observed.4. After treatment of Miao Nationality's herb, we research the changes of the indexes mentioned above.Results:1. after two months smoking and inhalation of lipopolysaccharide,the pathologic changes of COPD were appeared in lung tissues, which include Integration of the alveolar, the smooth muscle of small bronchial proliferated, a large number of lymphocytes and neutrophil infiltration and the enlargement of airspaces distal to the terminal bronchiole,the destruction and loss of the alveolar walls the mean lung internal lining interval (MLI) was significantly increased compared with the control group, the mean alveoli number (MAN) was significantly lower than control group, and there is a difference between COPD group and Miao Nationality s herb group (P<0.01); The WT% and WA% of COPD group are higher than control group(p<0.01). 2. Caspase-3 protein selectively expresses in alveolar and interstitial cells. The positive expressive rate of caspase-3 protein in COPD group is higher than control group (P<0.05), and the expression of caspase-3 mRNA is significantly higher than control group (P<0.01). There is a significantly positive relativity in the group between the expression of caspase-3 mRNA and MLI and a negtive relativity between the expression of caspase-3 mRNA and MAN(p<0.05).3. Survivin protein selectively expresses in smooth muscle of bronchioles and small vasculars; In COPD group and NC group, the positive rate of survivin proteins were 100% and 0% respectively,there is a significant different between two groups(P<0.05). There is a significantly positive relativity between the expression of survivin mRNA and WT% and WA% (p<0.05).4. In Miao Nationality's' herb group, the mean alveoli number (MAN) was significantly increased compared with COPD group(P<0.01), the mean lung internal lining interval (MLI) was significantly lower than COPD group(P<0.01). The expression of caspase-3 and survivin protein in Miao Nationality's' herb group were lower than COPD group(P<0.05) and the expression of caspase-3 and survivin mRNA were significantly lower than COPD group (P<0.01). WT% and WA% of Miao Nationality's' herb group were lower than COPD group(P<0.05;P<0.01).5. In dexamethasone group,the mean alveoli number (MAN) was significantly increased compared with COPD group(P<0.01), the mean lung internal lining interval (MLI) was significantly lower than COPD group(P<0.01). The expression of caspase-3 and survivin protein in dexamethasone group were significantly lower than COPD group(P<0.05) and the expression of caspase-3 and survivin mRNA were significantly lower than COPD group(P<0.01). WT% and WA% of dexamethasone group were lower than COPD group(P<0.05;P<0.01).
