Objectives:The purpose of the study was to detect the expressions of SNCG (γ-synuclein) and MAP2 (microtubule-associated protein-2) in gastric adenocarcinoma, to evaluate their roles in carcinogenesis, development, infiltration and metastasis as well as their clinical significances, to provide new axiliarily diagnostic marker for early finding and new target spots for targeted therapy.Methods:l.The expressions of SNCG and MAP2 were detected by SP immunohistochemical method in 90 cases of gastric adenocarcinoma and 40 cases of normal gastric mucosa.2.The expressions of SNCGmRNA and MAP2mRNA were detected by RT-PCR method in 29 cases of gastric adenocarcinoma,15 cases of normal gastric mucosa.Results:1.the results of immunohistochemistry The positive expressions of SNCG and MAP2 proteins in the gastric adenocarcinoma groups both were obviously higher than those in the normal gastric mucosa groups,and their differences were significant (P<0.01, P<0.01). The positive expressions of SNCG and MAP2 were both closely correlated with the depth of tumor infiltration and the metastasis of lymph node:the positive expressions of SNCG and MAP2 in invasion to chorion were higher than those invasion to mucosa or muscular layer(P<0.01,P<0.05); the positive expressions of SNCG and MAP2 in gastric adneocarcinoma with lymphnode metastasis were obviously higher than those without lymphnode metastasis (P<0.01, P<0.01); But both of them were not related to gender, age of patients and differentiation of carcinoma (P>0.05).2. the results of RT-PCR The expressional quantities of SNCGmRNA, MAP2mRNA in gastric adenocarcinoma both were higher than those in the normal gastric mucosa (P<0.01, P<0.05). The expressional quantities of SNCGmRNA and MAP2mRNA both were closely correlated with the depth of tumor infiltration and metastasis of lymph node:the expressional quantities of SNCGmRNA and MAP2mRNA in invasion to chorion were higher than those invasion to mucosa or muscular layer(P<0.05,P<0.05);the expressional quantities of SNCGmRNA and MAP2mRNA in gastric adenocar-cinoma with lymphnode metastasis were higher than those without lymphnode metastasis (P<0.01, P<0.05). And the expressional quantities of MAP2mRNA in gastric adenocarcinoma were related to gender (P<0.05). But the expressional quantities of SNCGmRNA in gastric adenocarcinoma were not related to gender (P>0.05).And both of them were not related to age of patients and differentiation of cancer (P>0.05).3.Correlation analysis (1) The positive expression of SNCG protein and expressional quantity of SNCGmRNA are positively correlated in the 29 cases of gastric adenocarcinoma (r=0.496, P<0.01). (2) The positive expression of MAP2 protein and expressional quantity of MAP2mRNA are positively correlated in the 29 cases of gastric adenocarcinoma (r=0.559, P<0.05). (3) The positive expressions of SNCG and MAP2 proteins are positively correlated in the 90 cases of gastric adenocarcinoma (r=0.620, P<0.01). (4) The expressional quantity of SNCGmRNA and MAP2mRNA are highly positively correlated in the 29 cases of gastric adenocarcinoma (r=0.432, P<0.05).Conlusion:1. SNCG and MAP2 may play an important role in carcinogenesis progression and metastasis of gastric adenocarcinoma.2. The expressions of SNCG and MAP2 protein in gastric adenocarcinoma were parallelly and positively correlated with the quantity of mRNA, either protein or mRNA detected by single or joint detection was representative.3.To detect the expressions of SNCG and MAP2 may provide guidance for the early finding, predicting prognosis of gastric adenocarcinoma.4. Further researches are needed to confirm whether SNCG or (/and) MAP2 can serve as target spots in the targeted therapy of gastric adenocarcinoma.5. The expression of MAP2mRNA in gastric adenocarcinoma is related to gender, whether the difference of physical characteristics or sample, needing further studies.
