Expression Of Transcription Factor T-bet In The Gastric Carcinoma Tissues And Its Correlation With Prognosis

Objective To analyze the expression of T-bet as a new transcription factor of T-box gene family in the gastric carcinoma tissues and its correlation with prognosis.Methods The number of T-bet positive lymphocyte in the surgical specimens of gastric cancer tissues were detected by using immunohistochemistry in 201 patients,data was analyzed by using SPSS 13.0.Results The number of T-bet positive lymphocyte in gastric carcinoma tissues was (15.2±9.5) /HPF.Single-factor analysis demonstrated that there was no statistical significance between the number of T-bet positive lymphocyte and sex,age,tumor site,histological type,recurrence,pathological grade or lymph node metastasis.However,there was a statistical significance between the number of T-bet positive lymphocyte and the tumor size,invasion depth or TNM staging.The number of T-bet positive lymphocyte was higher in the tumor diameter <5cm group (16.9±11.8)/HPF than in the tumor diameter≥5cm group (13.0±6.7)/HPF , the difference has statistical significance(t=2.35,P=0.0202). And the number of T-bet positive lymphocyte was obviously higher in the non-invasive group(20.2±14.6)/HPF than in the invasive group(14.1±8.3)/HPF , the difference has statistical significance(t=2.50, P=0.0138).Along with the increase of TNM staging,the number of T-bet positive lymphocyte has a gradually descendent trend , the difference has statistical significance(F=3.40,P=0.0196). Kaplan-Meier analytic result revealed that overall survival of the patients was longer in more infiltrative group(x≥19.15) than in lesser infiltrative group(x<19.15),the difference close to statistical level(χ2=1.90, P=0.168). The median survival time of the patients with more infiltrative group was significantly longer than those with lesser infiltrative group,The median survival time of the patients was 55.0 months (95%CI:28.2-81.8) in more infiltrative group and 37.0 months (95%CI:29.6-44.4) in lesser infiltrative group. Conclusions There was a correlation between the number of T-bet positive lymphocyte and the prognosis of gastric carcinoma,which play an important part in immunoregulation of gastric carcinoma tumor microenvironment.
