Objective:To explore the distribution and clinic significance of the new index on Anti-HBc and Anti-Hbe, which is made from detection of many times diluted Anti-HBc reactive samples and Anti-HBe reactive samples by chemiluminescent microparticle immunoassay(CMIA).Methods:All the Anti-HBc reactive samples and Anti-HBe reactive samples were devided into HBsAg and Anti-HBs reactive group. Each samle was diluted many times, Anti-HBc and Anti-HBe were detected in the Anti-HBc and Anti-HBe reactive samples. After each dilution, a detection curve graph was drew, the y axis represents the S/CO values measured by the Architect CMIA, and the x axis represents the logarithm of diluted times. A new index calculated from the area under each curve was to compare the two Anti-HBc and Anti-HBe reactive group. The S/CO value was also comparatively analysed in the two Anti-HBc and Anti-HBe reactive group. The correlations between the area value, ALT and HBV-DNA were analysed. Results:The difference area value between the two group of the Anti-HBc reactive samples was more significant(P<0.0001) than the S/CO value(P=0.013). The area value of the Anti-HBe reactive samples was significantly different between the two group(P<0.0001), but the S/CO value was not different(P=0.669>0.05). The area value on Anti-HBc and Anti-HBe were correlated with neither ALT(P>0.05), nor HBV-DNA(P>0.05). Our result showed that the area value calculated from the precise measurement with double dilution method is superior to the S/CO value in the classical measurement. Conclusion:The area value as the activity on Anti-HBc and Anti-HBe can obviously show the changes between onset phase and convalescent phase of chronic hepatitis B. It provides a valuable method for the diagnosis and treatment of chronic hepatitis B.
