Objective:This study by establishing the model of dog avascular necrosis of the necrotic bone repair under different stress, detect reconstruction of the expression level of BMP-2 and explore the BMP-2 under different stress conditions in osteonecrosis repair.Methods:Taking 28 healthy Beagle dogs.They were randomly divided into four groups: A, B and C for the experimental group,each of them have 8 dogs. The last 4 dogs are for the control group D.The femoral heads of the Beagle were surgically exposed, Resecting the anterolateral joint capsule and sutured, General stress caused by stimulation of the femoral head necrosis.Experimental group (24dogs):Dogs in each group were established model of femoral head necrosis, femoral head necrosis caused by different stress stimulation. Kill half of the dogs after 4 and 8 weeks respectively.Bilateral hip joint specimens were cut along the median sagittal saw and then detect the level of BMP-2 gene. According to Immunohistochemical area under the integrated optical density,we calculate in the tissue content of the hip of BMP-2.Results:It show that the expression of BMP-2 in 4 weeks and 8 weeks after surgery was significantly higher under Immunohistochemical pathology of the experimental groups.The absorbance of BMP-2 per unit area in each experimental group was higher than the controlled group in 4 weeks.And in 8 weeks, all experimental groups increased further, which medium-stress group increased most significantly. Compared with the other groups medium-stress group was significantly different(P<0.05)。Conclusion:BMP-2 of necrotic bone rebuilt express differently under different stress.And may play an important role in bone reconstruction.Increasing in femoral head of local BMP-2 expression may contribute to the repairation of avascular necrosis...
