Objective:To observe the change of Toll like receptors in the serum drawing from rabbits which have treated with recombinant human interleukin-10 after allogeneic skin graft, and explore the mechanism of recombinant human interleukin-10 resisting the immunological rejection, to provide laboratory evidence for the recombinant human interleukin-10's clinic utilization.Methods:1.18 rabbits as receptors,18 rabbits as donors to establish animal models for rabbit skin graft. The receptors are divided into 6 groups, so there are three rabbits in one group.①group:rhIL-10(5ug/kg);②group:rhIL-10(10ug/kg);③group:CsA(5mg/kg);④group:CsA(10mg/kg);⑤group:rhIL-10(5ug/kg)+CsA(5mg/kg);⑥group:normal saline.2. Give the medicine to rabbits which are skin graft receptors on the preoperative one day and consecutive 10 days by intramscular injection.3. Separate serum from the skin graf rabbits' blood on the preoperative one day, the first day, the fourth day, the seventh day, the fourteenth day, the twenty first day and the twenty eighth day after transplation. Then test TLR4, TLR7, TLR8, TLR9 using ELISA.Result:1. The results of TLR4 test show that:the serum TLR4 increases after skin graft compared with preoperative and reaches the peak concentration on the fourteenth day then decreases gradually in all groups. On the fourth day after transplation,⑥group is higher than⑤group, there is significant difference (p<0.05);①group is higher than⑤guoup, there is significant difference (p<0.05)2. The results of TLR7 test show that:the serum TLR7 increases after skin graft compared with preoperative and reaches the peak concentration on the fourteenth day then decrease gradually in all groups. On the seventh day after transplation,⑥group is higher than other five groups, there is significant difference (p<0.05)3. The results of TLR8 test show that:the serum TLR8 increases after skin graft compared with preoperative and reaches the peak concentration on the fourteenth day then decrease gradually in all groups. On the seventh day after transplation,⑥group is higher than②group, there is significant difference (p<0.05);⑥group is higher than④group, there is significant difference (p<0.05)4. The results of TLR9 test show that:the serum TLR9 increases after skin graft compared with preoperative and reaches the peak concentration on the fourteenth day then decrease gradually in all groups, but there is no significant difference in every two group.Conclusion:1. The serum TLR4, TLR7, TLR8, TLR9 increase obviously after the rabbit skin graft, which means that TLRs may participate in the allograft rejection by means of generous secreting which can mediate the transplantation inflammation reaction and promote the secretion of inflammation cytokines.2. As innate immunity molecular, the serum TLRs increase during the immune response and reach the peak concentration when the skin graft rejection, then decrease gradually, which means innate immunity participate in allograft rejection.3. TLRs participate in allograft rejection, so we can intervene in the TLRs signal pathway, which may be another way to control allograft rejection after organization transplantation.
