Objective:monitoring the change of CC,IL-8/CXCL8 chemokines in the serum drawing from rabbits through treated with rhIL-10 after skin transplantation to study the mechanism of rhIL-10 resisting the immune rejection and to provide the laboratory evidence for the rhIL-10 clinic utilization.Methods:(1) separating the skin transplantation rabbits to six groups randomly.①rhIL-10 low dose group.②rhIL-10 high dose group.③CsA low dose group.④CsA high dose group.⑤rhIL-10 and CsA low dose joint group.⑥normal saline group. (2) Mediation planning:giving the medicine the previous day of operation and consecutive 10 days by intramuscular injection. (3) We drew blood from every rabbit on the previous day of operation and the first day, the fourth day, the seventh day, the fourteenth day, twenty first day and twenty eighth day of after skin transplantation and separated the serum then tested the CC,IL-8/CXCL8 chemokine family with ELISA.Result:1. the results showed that the IL-8/CXCL8 serum levels in all groups were significantly higher after the skin transplantation than the preoperation (p<0.05);The peak of group⑥reached on the 7th day and maintained to the 14th day, while other groups reached the peak concentration on the 14th day then decreased gradually,1th day after surgery, the level of IL-8 was higher in group⑥than that in other groups, and there are significant difference between group⑥and group①,as well as group⑥and group②(p<0.05).4th day after surgery, IL-8 serum level was significantly higher in group⑥than that in all other groups except group⑤(p<0.05);besides, IL-8 level in group①was striking higher than that in group②,and IL-8 level in group①was striking higher than that in group②.7th day after surgery, he IL-8 serum levels in group⑥was higher than in all other groups with statistical difference (p<0.05).2. Compared with preoperation, the MCP-1/CCL2 serum levels in all groups were significantly increased (p<0.05), and their peak levels occurred on the 14th day, then decreased gradually.1th day and 4th day after surgery, MCP-1 level in group⑥was higher than any other groups; Compared with the negative control group, the MCP-1 serum levels in group①and group②as well as CsA positive control group were significantly lower (p<0.05); MCP-1 level in group②was striking higher than that in group④(p<0.05) 7th day after surgery, MCP-1 levels in group①and group⑤were lower than that in group⑥with statistical difference (p<0.05).3. The MIP-1α/CCL3 serum levels in all groups were significantly higher after the skin transplantation than the preoperation (p<0.05), and their peak levels occurred on the 14th day, then decreased gradually.4th day after surgery, the level of MIP-1αin group⑥was significantly higher than that in other groups (p<0.05); the level of MIP-1αin group②group③\group⑤\were significantly higher than that in group④(p<0.05).7th day after surgery, the level of MIP-1αin group⑥was significantly higher than that in other groups (p<0.05),and the level of MIP-1αin group②was significantly higher than that in group④(p<0.05)4. The MIP-1β/CCL4 serum levels in all groups were significantly higher after the skin transplantation than the preoperation (p<0.05), and their peak levels occurred on the 14th day, then decreased gradually.1th day after surgery, the MIP-1βserum level in group①was lower than that in group⑥and group⑤,and the MIP-1βserum level in group②was lower than that in group⑤with statistical difference (p<0.05).Compared with each other groups, there were no significant difference during 4~14d postoperation (p>0.05)Conclusion:1.During acute skin allograft rejection,the postive expression rate and expression inrensity of CC,IL-8/CXCL-8chemokines in all groups are significantly higher than preoperation,which hinted that these chemokines production incrase remarkably during acute rejection.It means that CC,IL-8/CXCL-8chemokines maybe paly a important role in acute rejection.2.There is a close correlation of CC,IL-8/CXCL8 chemokines levels with ischemia-reperfusion injury and with the acute allograft rejection, and they could be regarded as important sings of acute allograft rejection; 3.IL-8 and MCP-1 serum levels in rhIL-10 inhibition group was striking lower than that in negative control group, it means that they participate the transplantation rejection in an early phase as members of innate immunity.
